Targeting the DNA Damage Response in Cancer

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Targeting the DNA damage response (DDR) reshapes both tumor vulnerability and antitumor immunity. DNA lesions and DDR inhibitors can promote cytosolic nucleic‐acid sensing, antigen presentation, and immune activation, but can also induce adaptive immune escape.

Bruton's Tyrosine Kinase Inhibitors: Mechanisms, Efficacy, Toxicities and Applications for the Treatment of Human Diseases

open access: yesMedComm, Volume 7, Issue 10, October 2026.
BTK inhibitors suppress malignant B‐cell signaling while simultaneously reshaping immune regulation through effects on T cells and the tumor microenvironment. These mechanisms support combination immunotherapy strategies and also underlie the expanding application of BTK inhibitors in solid tumors, autoimmune and inflammatory diseases.

Alveolar Macrophages: Development, Respiratory Homeostasis, Signal Pathways, Diseases, and Therapeutic Opportunities

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Alveolar macrophages: development, respiratory homeostasis, signal pathways, diseases, and therapeutic opportunities: During embryonic development, fetal progenitors migrate to the developing lungs via a CX3CR1‐dependent mechanism to establish the TrAM pool.

Radiotherapy‐Induced Side Effects: Epidemiology, Pathogenesis, Prevention and Treatments

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Radiotherapy for malignancies frequently injures the organs’ systems through oxidative stress, fibrosis, inflammation, and vascular damage. This review systematically maps the epidemiology, pathogenesis, and emerging therapies to propose a mechanism‑network‑targeted framework for integrated, personalized care in radiation oncology.

Prodh2‐Mediated Mitochondrial Stress Drives TNF‐α‐Induced Myoblast Dysfunction and Sarcopenia in COPD

open access: yesAging Cell, Volume 25, Issue 10, October 2026.
TNF‐α upregulates Prodh2 via p53 in COPD myoblasts, driving mitochondrial ROS–induced cGAS/STING activation and myoblast dysfunction. This metabolic‐immune pathway underlies sarcopenia, positioning Prodh2 as a therapeutic target for COPD‐associated muscle wasting. ABSTRACT Sarcopenia in chronic obstructive pulmonary disease (COPD) is driven by elevated
