Adaptation of Echinococcus Multilocularis to Oxidative Stress Depends on EmPDK‐Mediated Metabolic Reprogramming

open access: yesAdvanced Science, EarlyView.
Metabolic adaption presents a vital survival strategy for E. multilocularis. This study identifies EmPDK as a central metabolic regulator governing glucose metabolic reprogramming. Oxidative stress activates EmHIF1α to induce EmPDK expression. Mechanistic characterization reveals this ROS/EmHIF1α/EmPDK axis drives glycolytic reprogramming to sustain ...

USP5 Stabilizes TGFBR1 to Drive Vascular Smooth Muscle Cell Senescence and Atherosclerosis

open access: yesAdvanced Science, EarlyView.
This study reveals that USP5 drives vascular smooth muscle cell senescence and atherosclerosis by stabilizing TGFBR1, suppressing IDH2, and promoting glycolytic reprogramming, identifying the USP5‐TGFBR1‐IDH2 axis as a potential therapeutic target. ABSTRACT Vascular smooth muscle cell (VSMC) senescence contributes importantly to atherosclerotic plaque ...

Redefining care for IDH-mutant low-grade gliomas: Clinical implications of the INDIGO trial and updated guidelines

open access: yesClinical Medicine
Pathogenic variants in the genes encoding isocitrate dehydrogenase (IDH1 and IDH2) are the defining driver mutations in diffuse low-grade gliomas. When IDH is mutated, the production of the oncometabolite 2-hydroxyglutarate triggers downstream metabolic ...

Isocitrate dehydrogenase mutation hot spots in acute lymphoblastic leukemia and oral cancer

open access: yesKaohsiung Journal of Medical Sciences, 2012
Isocitrate dehydrogenase (IDH) encodes a nicotinamide adenine dinucleotide phosphate+-dependent enzyme for oxidative decarboxylation of isocitrate and has an essential role in the tricarboxylic acid cycle.

Molecular Aberrations Stratify Grade 2 Astrocytomas Into Several Rare Entities: Prognostic and Therapeutic Implications

open access: yesFrontiers in Oncology, 2022
The identification of specific molecular aberrations guides the prognostic stratification and management of grade 2 astrocytomas. Mutations in isocitrate dehydrogenase (IDH) 1 and 2, found in the majority of adult diffuse low-grade glioma (DLGG), seem to

EBV‐Driven ODC1 Upregulation Enhances Polyamine Anabolism to Promote Viral Replication and Cisplatin Resistance in Nasopharyngeal Carcinoma

open access: yesAdvanced Science, EarlyView.
EBV‑BZLF1 initiates ODC1‑driven polyamine anabolism that correlates with poor clinical prognosis in nasopharyngeal carcinoma. The ODC1–spermidine axis promotes viral replication, cell proliferation and innate immune evasion. Pharmacological inhibition of ODC1 rescues cisplatin sensitivity, establishing ODC1 as a viable therapeutic target for EBV ...

A comparison of the phosphorylated and unphosphorylated forms of isocitrate dehydrogenase from Escherichia coli ML308 [PDF]

open access: yes, 1984
NADP+ can protect active isocitrate dehydrogenase against attack by several proteases. Inactive phosphorylated isocitrate dehydrogenase is much less susceptible to proteolysis than the active enzyme, and it is not protected by NADP+.

The Cancer Cell Metabolic Reprogramming Remodels the Tumor Microenvironment: Molecular Mechanisms and Therapeutic Strategies

open access: yesAdvanced Science, EarlyView.
This review elucidates how cancer cell metabolic reprogramming—across glucose, lipid, amino acid, and nucleotide pathways—remodels the tumor microenvironment to suppress anti‐tumor immunity and promote immune escape. Targeting these metabolic axes offers promising strategies to overcome immunotherapy resistance and enhance cancer treatment.

Mutagenesis and Laue Structures of Enzyme Intermediates: Isocitrate Dehydrogenase [PDF]

open access: yes, 1995
Site-directed mutagenesis and Laue diffraction data to 2.5 A resolution were used to solve the structures of two sequential intermediates formed during the catalytic actions of isocitrate dehydrogenase.

Targeting the KAT7/H3K14ac/RAC2 Axis Mediates OXPHOS to Promote Stemness Maintenance and Malignancy in Glioblastoma

open access: yesAdvanced Science, EarlyView.
SOX2 upregulates KAT7 in glioma stem cells, where KAT7 deposits H3K14 acetylation at the RAC2 promoter to activate RAC2 transcription. RAC2‐GTP subsequently promotes PAK1/2/3 phosphorylation, increasing tricarboxylic acid cycle flux and mitochondrial respiration to support glioblastoma malignancy.
