Gαi1/3 Is a Novel Regulatory Target for RANKL Signal Transduction and Osteoporosis

open access: yesAdvanced Science, EarlyView.
ABSTRACT Osteoporosis, characterized by progressive bone loss and increased fracture risk, is a growing concern as the population ages. Current treatments, though advanced, remain limited, underscoring the necessity for novel therapeutic targets. Recent studies have shown that the immune system plays a key role in osteoporosis, with osteoclasts driving

Oxidative stress-induced apoptosis in granulosa cells involves JNK, p53 and Puma

open access: yesOncoTarget, 2017
Reactive oxygen species (ROS) play important roles in follicular development and survival. Granulosa cell death is associated with increased ROS, but the mechanism of granulosa cell death induced by ROS is not clear. In order to define the molecular link

The JNK-interacting Protein-1 Scaffold Protein Targets MAPK Phosphatase-7 to Dephosphorylate JNK [PDF]

open access: yesJournal of Biological Chemistry, 2003
The c-Jun N-terminal kinase (JNK) group of mitogen-activated protein kinases (MAPKs) are activated by pleiotropic signals including environmental stresses, growth factors, and hormones. A subset of JNK can bind to distinct scaffold proteins that also bind upstream kinases of the JNK pathway, allowing sequential kinase activation within a signaling ...

CDK4/6 Inhibition Induces CD8+ T Cell Antitumor Immunity via MIF‐Induced Functional Orchestration of Tumor‐Associated Macrophages

open access: yesAdvanced Science, EarlyView.
CDK4/6 inhibition promotes CD8+ T cell expansion through tumor‐macrophage crosstalk by activating HIF‐1α and enhancing MIF‐CD44/CD74 signaling. This reprograms TAMs to boost MHC‐I antigen presentation, and CDK4/6 inhibitor‐trained M1 TAM supernatant therapy synergizes with low‐dose PD‐1 blockade to restore antitumor immunity.

Beyond antioxidants: the cellular and molecular interactions of flavonoids and how these underpin their actions on the brain [PDF]

open access: yes, 2010
The consumption of flavonoid-rich foods and beverages has been suggested to limit the neurodegeneration associated with a variety of neurological disorders and to prevent or reverse normal or abnormal deteriorations in cognitive performance.

A sestrin-dependent Erk/Jnk/p38 MAPK activation complex inhibits immunity during ageing

open access: yesNature Immunology, 2016
Mitogen-activated protein kinases (MAPKs) including Erk, Jnk and p38 regulate diverse cellular functions and are thought to be controlled by independent upstream activation cascades. Here we show that the sestrins bind to and coordinate simultaneous Erk,

Notch2 Directs aTreg Cell Fate toward Immunoregulation or Inflammatory Pyroptosis

open access: yesAdvanced Science, EarlyView.
Schematic illustration showing that the Notch2 intracellular domain (NICD2) facilitates pyroptosis resistance in activated Tregs through the RREB1/Foxo1 signaling pathway. In addition, Notch2‐mediated pyroptosis resistance in activated Tregs promotes immunoregulatory capacity, thereby attenuating Th2‐driven inflammatory responses in allergic rhinitis ...

TNF receptor–related factor 3 inactivation promotes the development of intrahepatic cholangiocarcinoma through NF‐κB‐inducing kinase–mediated hepatocyte transdifferentiation

open access: yesHepatology, EarlyView., 2022
Abstract Background and Aims Intrahepatic cholangiocarcinoma (ICC) is a deadly but poorly understood disease, and its treatment options are very limited. The aim of this study was to identify the molecular drivers of ICC and search for therapeutic targets.

JNK regulates muscle remodeling via myostatin/SMAD inhibition

open access: yesNature Communications, 2018
Skeletal muscle has a remarkable plasticity to adapt and remodel in response to environmental cues, such as physical exercise. Endurance exercise stimulates improvements in muscle oxidative capacity, while resistance exercise induces muscle growth.

JNK inhibition arrests cotranslational degradation

open access: yesBiochimica et Biophysica Acta (BBA) - Molecular Cell Research, 2010
Adhesion to fibronectin stimulates protein synthesis (translation) of fibroblasts. Protein synthesis stimulation is dependent from the activation of beta(1)-integrin. beta(1)-Integrin elicits a PI3K cascade that modulates eIF4F (eukaryotic initiation factor 4F) complex formation. In the attempt to further dissect elements of the PI3K cascade that might
