De-escalated therapy for HR+/HER2+ breast cancer patients with Ki67 response after 2-week letrozole: results of the PerELISA neoadjuvant study

open access: yesAnnals of Oncology, 2019
Background In human epidermal growth factor receptor 2 (HER2+) breast cancers, neoadjuvant trials of chemotherapy plus anti-HER2 treatment consistently showed lower pathologic complete response (pCR) rates in hormone receptor (HR) positive versus ...

Ki67 Immunofluorescence on Bovine Cell Lines

open access: yesBIO-PROTOCOL, 2013
This is a rapid protocol to test the effects of drugs treatment on bovine cell replication using Ki67 staining. Ki67 is associated with cell proliferation and is present during all active phases of the cell cycle (G1, S, G2, and mitosis), but is absent from resting cells (G0).

3D Soft Hydrogels Induce Human Mesenchymal Stem Cells “Deep” Quiescence

open access: yesAdvanced Healthcare Materials, EarlyView.
Three‐dimensional soft hydrogels mimicking the bone marrow niche induce deep quiescence in human mesenchymal stem cells. Unlike 2D culture, 3D matrices halt proliferation, regulate cell‐cycle and quiescence markers, and downregulate mTORC1 signaling, preserving stem cell phenotype and therapeutic potential ex vivo.

Combined quantitative measures of ER, PR, HER2, and KI67 provide more prognostic information than categorical combinations in luminal breast cancer

open access: yesModern Pathology, 2019
Although most women with luminal breast cancer do well on endocrine therapy alone, some will develop fatal recurrence thereby necessitating the need to prospectively determine those for whom additional cytotoxic therapy will be beneficial.

Architecture of an All‐In‐One Microfluidic Platform for Accelerated Cancer Seeding, Enhanced Spheroid Formation, and Dynamic Drug Screening Trials

open access: yesAdvanced Materials Technologies, EarlyView.
An all‐in‐one microfluidic platform consists of two‐layered PDMS channels integrated with a hydrogel microwell array. This platform enables flow‐driven tumor cell capture, enhanced spheroid formation within the microwells, and advective drug testing. ABSTRACT Effective cancer drug screening requires platforms that replicate the physiological complexity

DRD2 Deficiency Underlies Pituitary Adenoma Dependent on Escherichia coli Translocation from the Gut

open access: yesAdvanced Science, EarlyView.
Dopamine receptor D2 (DRD2) deficiency disrupts epithelial barrier integrity in both the gut and pituitary gland, facilitating translocation of gut‐derived Escherichia coli into pituitary tissue. Intratumoral E. coli are phagocytosed by microglia, triggering GSDMD‐dependent pyroptosis and HMGB1 release, which subsequently activates MAPK signaling to ...

Pre‐Encoded IFN‐I Sensitivity Exacerbates Memory T Cell Senescence in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Type I interferon (IFN‐I) signaling promotes p21‐dependent cell cycle arrest in senescent tumor‐specific memory T cells, resulting in poor proliferative responses and solid tumor regression during cancer vaccination. Conversely, IFNα/β receptor blockade reinvigorates T cell proliferation to regress solid tumors and is more effective with increasing ...

FAPα+ Macrophages Orchestrate Immune Evasion in Multiple Myeloma by Dual Regulation of PD‐L1 and T Cell Senescence

open access: yesAdvanced Science, EarlyView.
Fibroblast activation protein alpha‐positive (FAPα+) macrophages, a distinct subset of multiple myeloma (MM)‐associated macrophages, drive immune evasion in MM through multi‐faceted mechanisms. FAPα physically interacts with vimentin (VIM) and triggers its phosphorylation at the S72 residue, which in turn induces PD‐L1 transcription. Additionally, FAPα

An Extended Culture System that Supports Human Primordial Germ Cell-like Cell Survival and Initiation of DNA Methylation Erasure. [PDF]

open access: yes, 2020
The development of an in vitro system in which human primordial germ cell-like cells (hPGCLCs) are generated from human pluripotent stem cells (hPSCs) has been invaluable to further our understanding of human primordial germ cell (hPGC) specification ...
