Diltiazem N‐oxide, a novel ischaemia‐activated prodrug, prevents ischaemia‐induced ventricular fibrillation without off‐target adverse effects

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose Ventricular fibrillation (VF) in coronary heart disease accounts for up to 70% of sudden cardiac death. We examined whether diltiazem N‐oxide (DNO) has ischaemia‐selective antiarrhythmic activity. Experimental Approach Randomised and blinded experiments were performed in rat isolated hearts and in anaesthetised rats to ...

Toll‐like receptor 4 mediates lipopolysaccharide‐induced emesis

open access: yesBritish Journal of Pharmacology, EarlyView.
Background and Purpose Lipopolysaccharide (LPS) induces inflammation and sepsis through Toll‐like receptor 4 (TLR4) activation. Common laboratory animals do not exhibit emesis, but administration of LPS in piglets, cats, and dogs induces emesis via peripheral mechanisms.

The Key Enzymes of Carbon Metabolism and the Glutathione Antioxidant System Protect Yarrowia lipolytica Yeast Against pH-Induced Stress

open access: yesJournal of Fungi
In this study, we first thoroughly assayed the response of the key enzymes of energy metabolism and the antioxidant system in Yarrowia lipolytica yeast at extreme pH.

Primordial Krebs-cycle-like non-enzymatic reactions detected by mass spectrometry and nuclear magnetic resonance

open access: yes, 2017
Background: Metabolism is the process of nutrient uptake and conversion, and executed by the metabolic network. Its evolutionary precursors most likely originated in non-enzymatic chemistry. To be exploitable in a Darwinian process that forms a metabolic

Gβγ subunit activation of KV7 and BKCa channels underlies calcitonin gene‐related peptide vasorelaxation in myogenic rat coronary resistance arteries

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose CGRP is a potent, clinically relevant coronary vasodilator known to play a role in cardioprotection. Here, we investigated the precise intracellular signalling pathways leading to vasodilation in small coronary arteries.

Presence of Krebs Cycle Intermediates in Primary Mycelia of an Actinomycete [PDF]

open access: yes, 1968
The purpose of this study is to determine the presence or absence of a typical Krebs cycle metabolism in the primary mycelia of an aquatic ...

AMPK‐mediated prevention of vascular dysfunction with metformin: Experimental and population‐based evidence

open access: yesBritish Journal of Pharmacology, EarlyView.
Angiotensin II induces vascular inflammation, senescence, extracellular matrix (ECM) degradation and endothelial dysfunction, thus impairing SK+/IK+ channel function and reducing endothelial‐dependent hyperpolarisation (EDH). Metformin activates AMPK, increasing SK+/IK+ channel function and restoring vasorelaxation.

Diagnostic Yield and Clinical Impact of Comprehensive WES/WGS Testing Beyond Common Genetic Causes in Hereditary Optic Atrophy

open access: yesClinical Genetics, EarlyView.
Opticus atrophy—Genetic testing with WES/WGS in 62 patients with optic atrophy provided a genetic diagnosis in 21 patients (33.9%). 42.9% of these involved non‐OPA1 genes, including WFS1, ACO2, NR2F1, UCHL1, CACNA1F, and COQ2, where the genetic diagnosis prompted additional clinical evaluation, surveillance, or therapeutic intervention.
