Optimization of B Cell Responses in Human Immune System Mice Through Organoid Based Screening
Human immune system (HIS) mice show limited B cell responses to immunization. Using HIS mouse spleen organoids, this study identifies cytokine combinations and innate immune signals that drive human B cell expansion, differentiation, and class‐switch. Applying these signals in vivo through temporal separation enables antigen‐specific IgG responses and ...

ABSTRACT Iron overload is a common metabolic disturbance in cancer and contributes to poor outcomes in renal cell carcinoma (RCC), yet its effects on the tumour immune microenvironment remain unclear. Here we identify a previously unrecognized immunosuppressive axis in which iron overload downregulates the palmitoyltransferase ZDHHC12 in CD8+ T cells ...

Palmitic Acid Promotes Antiviral Innate Immunity via ZDHHC20‐Mediated CMPK2 Palmitoylation
Metabolites have important functions in innate immune activation and regulation. Wang et al. uncover metabolic regulation of antiviral immunity through CMPK2 palmitoylation, which regulates CMPK2 mitochondrial localization and is promoted by ZDHHC20 but reversed by PPT1, inhibition of which antagonizes viral infection in mice.

Immunoinformatics Approach to T-cell Epitopes in Human Immunodeficiency Virus
Human immunodeficiency virus (HIV) is major human disease which belongs to lentivirus (retrovirus). It is a major cause of acquired immunodeficiency syndrome (AIDS), a condition in humans in which progressive failure of the immune system allows life ...

Enhanced Intracellular Stability and Translation Efficiency of mRNA Drugs by a 2‐arm mRNA Platform
We constructed a 2‐arm mRNA, characterized by a unique topology formed through the dimerization of two mRNA 3’ tails. The 2‐arm mRNA improves 3’ tail stability and resistance to nuclease degradation, resulting in an intracellular half‐life of up to 65 h. This method substantially enhances the translation capacity of mRNA drugs.

Construction of a Vero cell line expression human KREMEN1 for the development of CVA6 vaccines
Coxsackievirus A6 (CVA6) has emerged as a major pathogen causing hand, foot and mouth disease (HFMD) outbreaks worldwide. The CVA6 epidemic poses a new challenge in HFMD control since there is currently no vaccine available against CVA6 infections.

High glucose triggers corneal endothelial dysfunction by impairing FOXO1‐mediated ITPR1 transcription, leading to disrupted mitochondria‐associated membrane (MAM) integrity and defective ER‐to‐mitochondria Ca2+ transfer. This study develops an innovative viscous DES‐AAV‐Foxo1 delivery system with enhanced transfection efficiency. This non‐invasive gene

WTAP drives cartilage regeneration by activating an LRP1‐dependent lipid metabolic program in macrophages, enhancing IL‐10 and TGF‐β secretion to promote chondrogenic differentiation. Leveraging this mechanism, virtual screening identifies LRP1‐targeting compounds that effectively stimulate cartilage repair, highlighting a druggable epigenetic ...

Sheep Lentivirus Infection in different production systems
Small ruminant lentiviruses (SRLV) are a group of viruses that infect and transmit among ovine species. SRLV infection inflict progressive and longstanding infections, the majority as asymptomatic or developing late onset clinical signs. SRLV infection is a serious problem on ovine production, not only for animals’ wellbeing as well to the herd ...
