Data mining reveals novel gene drivers of lenvatinib resistance in hepatocellular carcinoma

open access: yesAnnals of Hepatology
Introduction and Objectives: Liver cancer is the sixth most common malignancy and the third leading cause of cancer-related deaths globally. Hepatocellular carcinoma (HCC) is the most prevalent type, accounting for nearly 90 % of all liver cancer cases ...

Enhanced peripheral blood mononuclear cell lipid metabolism drives poor immunotherapy response via ferroptotic stress‐induced T cell exhaustion in hepatocellular carcinoma

open access: yesInterdisciplinary Medicine, EarlyView.
Integrated multi‐omics reveals that CD36‐mediated lipid overload in PBMCs drives T cell exhaustion via sub‐lethal ferroptotic stress and AKT suppression. This systemic metabolic signature serves as a robust predictor of immunotherapy response in HCC, suggesting that modulating lipid metabolism can rescue immune fitness.

Composite Inflammatory Score for Predicting Progression‐Free Survival in Metastatic Clear Cell Renal Cell Carcinoma Treated With First‐Line Toripalimab Plus Axitinib

open access: yesThe Kaohsiung Journal of Medical Sciences, EarlyView.
ABSTRACT Routine inflammatory and nutritional markers may help stratify progression risk in metastatic clear cell renal cell carcinoma during immunotherapy‐based treatment. This single‐center retrospective study included 237 patients who received first‐line toripalimab plus axitinib therapy.

The FGF/FGFR System in the Biology and Therapeutic Landscape of Pediatric CNS Tumors

open access: yesMedicinal Research Reviews, EarlyView.
ABSTRACT Central nervous system (CNS) tumors are the most common solid malignancies in children, comprising a highly heterogeneous group of neoplasms defined by distinct molecular alterations and clinical behaviors. Advances in molecular genetics have underscored the relevance of specific signaling pathways in driving pediatric tumorigenesis, among ...

Differential FGFR1 copy number status displays unique tumour immune microenvironment in triple‐negative breast cancer

open access: yesThe Journal of Pathology, EarlyView.
Abstract Fibroblast growth factor receptor 1 (FGFR1) amplification, a significant cancer alteration with both prognostic and therapeutic relevance, lacks a standardized definition. The relationship between FGFR1 copy number (CN) variations and tumour microenvironment (TME) characteristics also remains poorly understood.

Anti-cancer Effects of HNHA and Lenvatinib by the Suppression of EMT-Mediated Drug Resistance in Cancer Stem Cells [PDF]

open access: yes, 2018
Anaplastic thyroid cancer (ATC) constitutes less than 2% of total thyroid cancers but accounts for 20–40% of thyroid cancer-related deaths. Cancer stem cell drug resistance represents a primary factor hindering treatment.

PPM1G‒MET axis sustains AKT signaling to drive Twist‐associated EMT and drug resistance in hepatocellular carcinoma

open access: yesVIEW, EarlyView.
Hepatocellular carcinoma (HCC) is frequently resistant to targeted therapies. We identify protein phosphatase Mg2+/Mn2+‐dependent 1G (PPM1G) as a key phosphatase that stabilizes mesenchymal‐epithelial transition factor (MET) by inhibiting its ubiquitination, thereby sustaining protein kinase B (AKT) signaling and promoting Twist‐mediated epithelial ...

LINC00892 inhibits hepatocellular carcinoma progression through SBDS/EIF6‐mediated ribosomal stress and interacting with the immune microenvironment

open access: yesVIEW, EarlyView.
LINC00892 is lowly expressed in HCC. LINC00892 overexpression induces ribosomal stress via inhibiting SBDS, restrains EIF6 nuclear translocation and activates p53 pathway. LINC00892 overexpression and p53 activation jointly suppress HCC progression. High LINC00892 is associted with infiltration of CD8+ T cells, B cells and macrophages, and associated ...
