Engineered Mesenchymal Stem Cell–NK Cell Complexes for Spatially Targeted and Functionally Revitalized Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
IL‐15‐engineered stem cell–NK cell complexes, assembled via bioorthogonal chemistry, enable effective lung cancer immunotherapy. Abstract Natural killer (NK) cells represent a powerful immunotherapeutic strategy due to their intrinsic cytotoxicity and ability to target tumor cells independently of antigen presentation.

Comprehensive Profiling of N6‐methyladnosine (m6A) Readouts Reveals Novel m6A Readers That Regulate Human Embryonic Stem Cell Differentiation

open access: yesAdvanced Science, EarlyView.
This research deciphers the m6A transcriptome by profiling its sites and functional readout effects: from mRNA stability, translation to alternative splicing, across five different cell types. Machine learning model identifies novel m6A‐binding proteins DDX6 and FXR2 and novel m6A reader proteins FUBP3 and L1TD1.

Novel Vascularized Human Liver Organoids for Modeling Alcohol‐Induced Liver Injury and Developing Hepatoprotective Therapy

open access: yesAdvanced Science, EarlyView.
This study successfully engineered vascularized liver organoids (3HLOs) by co‐culturing human reprogrammed hepatocyte‐like cells (hrHLs) with human umbilical vein endothelial cells (HUVECs) and human umbilical mesenchymal stem cells (HUMSCs). Upon implantation, the 3HLOs established functional vascular anastomosis with the host circulation and ...

Aberrant SUMOylation Restricts the Targetable Cancer Immunopeptidome

open access: yesAdvanced Science, EarlyView.
Pharmacological SUMOylation inhibition (SUMOi) counteracts tumor immune evasion by unmasking an immunogenic HLA‐I peptide and neoepitope repertoire. By restoring HLA‐I ligand availability through increased antigen processing and presentation, enhanced proteasomal cleavage, and modulated TAP1 peptide affinity, SUMOi boosts tumor immunogenicity ...

Transplantation of GABAergic Interneuron Progenitors Restores Cortical Circuit Function in an Alzheimer's Disease Mouse Model

open access: yesAdvanced Science, EarlyView.
Transplantation of medial ganglionic eminence (MGE) interneuron progenitors into APP/PS1 cortices restored the slow oscillation characteristic of Alzheimer's disease. Donor cells survived, migrated, and matured into functional GABAergic interneurons, forming synaptic connections.

ETV1 Drives CD4+ T Cell‐Mediated Intestinal Inflammation in Inflammatory Bowel Disease Through Amino Acid Transporter Slc7a5

open access: yesAdvanced Science, EarlyView.
This study identifies the transcription factor ETV1 as a key driver of CD4⁺ T cell‐mediated intestinal inflammation in inflammatory bowel disease (IBD). ETV1 promotes CD4⁺ T cell activation, proliferation, and Th17 differentiation by activating the amino acid transporter SLC7A5, fueling metabolic reprogramming.
