Luciferase-Specific Coelenterazine Analogues for Optical Contamination-Free Bioassays

open access: yesScientific Reports, 2017
Spectral overlaps among the multiple optical readouts commonly cause optical contamination in fluorescence and bioluminescence. To tackle this issue, we created five-different lineages of coelenterazine (CTZ) analogues designed to selectively illuminate ...

rWTC‐MBTA Vaccine, Alone and Enhanced with Anti‐PD1, Elicits Immune Responses against CNS and Peripheral B‐Cell Lymphoma

open access: yesAdvanced Science, EarlyView.
An autologous whole‐tumor‐cell vaccine (rWTC‐MBTA) is evaluated in murine CNS lymphoma. Subcutaneous vaccination activates dendritic cells, broadens T‐cell priming, and drives lymphocyte trafficking to brain tumors, producing durable tumor control. Longitudinal bioluminescence and adoptive‐transfer assays verify CNS engagement. Combination with anti‐PD‐

ClpL Chaperone as a Possible Component of the Disaggregase Activity of Limosilactobacillus fermentum U-21

open access: yesBiology
The L. fermentum U-21 strain, known for secreting chaperones into the extracellular milieu, emerges as a promising candidate for the development of novel therapeutics termed disaggregases for Parkinson’s disease.

GATA4‐Driven Transcription of HtrA1 Promotes Cellular Senescence in Ménière's Disease and Age‐Related Audio‐Vestibular Dysfunction

open access: yesAdvanced Science, EarlyView.
This study identifies the HDAC6/GATA4/HtrA1 axis as a critical driver of cellular senescence in the inner ear. GATA4 nuclear translocation, facilitated by HDAC6 downregulation, transcriptionally activates HtrA1, promoting hair cell senescence, SASP, and audio‐vestibular dysfunction in models of Ménière's disease and age‐related audio‐vestibular ...

Interference of metal ions on the bioluminescent signal of firefly, Renilla, and NanoLuc luciferases in high-throughput screening assays

open access: yesFrontiers in Chemistry
Bioluminescent high-throughput screening (HTS) assays, based largely on the activity of firefly (FLuc), Renilla (RLuc), and/or NanoLuc (NLuc) luciferases, are widely utilised in research and drug discovery. In this study, we quantify the luciferase-based

A View on the Active Site of Firefly Luciferase

open access: yesNatural Product Communications, 2008
The bioluminescent enzyme firefly luciferase has been the subject of detailed biochemical studies since the late 1940s. These studies have culminated in the cloning and sequence determination of some 161 different c-DNAs and/or genes for firefly ...

Divergent Regulatory Effects of Jasmonic Acid on Tomato Lycopene Biosynthesis Under Light and Dark Conditions

open access: yesAdvanced Science, EarlyView.
This study reveals the mechanism by which jasmonic acid (JA) regulates lycopene synthesis under light and dark conditions. In light, JA activates SlMYC2, which suppresses SlPIF1a and promotes SlPSY1 expression. In darkness, JA induces the acetyltransferase SlNATA1, which acetylates the dark‐accumulated SlPIF1a, thereby repressing SlPSY1 expression ...

HSP90 inhibitors stimulate DNAJB4 protein expression through a mechanism involving N6-methyladenosine. [PDF]

open access: yes, 2019
Small-molecule inhibitors for the 90-kDa heat shock protein (HSP90) have been extensively exploited in preclinical studies for the therapeutic interventions of human diseases accompanied with proteotoxic stress.

Endocytic Control of Cell‐Autonomous and Non‐Cell‐Autonomous Functions of p53

open access: yesAdvanced Science, EarlyView.
NUMB Ex3‐containing isoforms localize to the plasma membrane, where they recruit p53 through SNX9 and direct it to multivesicular bodies and exosomes. Exported p53 is taken up by neighboring cells and activates nuclear programs, revealing an intercellular, exosome‐based pathway that might help establish a tumor‐suppressive microenvironment.

TEAD1 Enhances Exosome Secretion and Promotes Exosome‐Mediated Tissue Regeneration

open access: yesAdvanced Science, EarlyView.
TEAD1 functions as a crucial molecular switch regulating exosome secretion in various cell types. TEAD1 enhances exosome secretion by upregulating key proteins associated with exosome secretion, including RAB11, CD9, and SNAP23. This study reveals a novel role for TEAD1 in regulating exosome secretion and tissue regeneration, particularly in diabetic ...
