On the origins of enzyme inhibitor selectivity and promiscuity: a case study of protein kinase binding to staurosporine

open access: yes, 2009
Protein kinases are important regulatory enzymes in signal transduction and in cell regulation. Understanding inhibition mechanisms of kinases is important for the further development of new therapies for cancer and inflammatory diseases.

Hyperosmotic stress induces PARP1‐mediated HPF1‐dependent mono(ADP‐ribosyl)ation

open access: yesFEBS Letters, EarlyView.
Sorbitol‐induced hyperosmotic stress rapidly induces reversible mono(ADP‐ribosyl)ation (MARylation) on PARP1 without the signs of genotoxic signaling. We show that PARP1 autoMARylation is HPF1 dependent and forms hydroxylamine‐resistant O‐glycosidic linkages.

Bayesian analysis of dynamic phosphoproteomic data identifies protein kinases mediating GPCR responses

open access: yesCell Communication and Signaling, 2022
Background A major goal in the discovery of cellular signaling networks is to identify regulated phosphorylation sites (“phosphosites”) and map them to the responsible protein kinases.

MAP Kinase Pathways [PDF]

open access: yesCold Spring Harbor Perspectives in Biology, 2012
MAP kinases are activated within protein kinase cascades that regulate cell proliferation, differentiation, and death. In mammals, MAP kinases are grouped into three families: ERKs, JNKs, and p38/SAPKs.

An isoform of 14‐3‐3 protein regulates transbilayer lipid movement at the plasma membrane

open access: yesFEBS Letters, EarlyView.
Loss of 14‐3‐3ζ in CHO cells confers resistance to exogenous phosphatidylserine (PS) and impairs endocytosis‐independent inward flip‐flop of fluorescent PS at the plasma membrane. RNAi‐mediated knockdown reproduces this defect, while no additive effect is seen in ATP11C‐deficient cells.

RAF kinase activity regulates neuroepithelial cell proliferation and neuronal progenitor cell differentiation during early inner ear development

open access: yes, 2010
Background: Early inner ear development requires the strict regulation of cell proliferation, survival, migration and differentiation, coordinated by the concerted action of extrinsic and intrinsic factors.

Stress and Vascular Responses: Atheroprotective Effect of Laminar Fluid Shear Stress in Endothelial Cells: Possible Role of Mitogen-Activated Protein Kinases

open access: yesJournal of Pharmacological Sciences, 2003
Atherosclerosis preferentially occurs in areas of turbulent blood flow and low fluid shear stress, whereas laminar blood flow and high shear stress are atheroprotective.
