Anion channels for amino acids in MDCK cells

American Journal of Physiology-Cell Physiology, 1992
Large losses of amino acids by diffusion were previously observed in Madin-Darby canine kidney (MDCK) cells during volume regulation. Also, an outward rectifying anion channel was activated. Because this channel was not selective among anions, it was suggested that it could be permeable to amino acids.

Polarized exocytosis in mdck cells is regulated by phosphorylation

Journal of Cell Science, 1995
ABSTRACT Protein phosphorylation and dephosphorylation systems modulate many cellular activities and have recently been implicated in the in vitro transport of newly synthesized proteins. Here we show that polarized transport from the Golgi to the plasma membrane in intact MDCK cells is regulated by phosphorylation-dephosphorylation ...

Methods for Establishing Rab Knockout MDCK Cells

2021
The Rab family small GTPases are key regulators of intracellular membrane traffic that are conserved in all eukaryotic cells. Rabs are thought to regulate various steps of membrane traffic, including the budding, transport, tethering, docking, and fusion of vesicles or organelles.

Measuring receptor recycling in polarized MDCK cells

2015
Recycling of proteins such as channels, pumps, and receptors is critical for epithelial cell function. In this chapter we present a method to measure receptor recycling in polarized Madin-Darby canine kidney cells using an iodinated ligand. We describe a technique to iodinate transferrin (Tf), we discuss how (125)I-Tf can be used to label a cohort of ...

Ion Transport in MDCK Cells

1985
The use of monolayers of MDCK cells as a model system for natural transporting epithelia created the necessity of learning about their ion-translocating mechanisms. These mechanisms have been analyzed directly by measuring unidirectional fluxes of tracers, and indirectly through their consequences on electrical parameters and blister-forming activity ...

Transfection Alters Ion Transport in MDCK Cells

Journal of Membrane Biology, 1996
In the course of an investigation into the effect of Tamm-Horsfall protein (THP) on ion transport, we performed stable transfection of THP into MDCK cells using the SV40 or the cytomegalovirus (CMV) promoter. As controls, we transfected MDCK cells with an "empty" plasmid containing SV40 or CMV promoter but without THP cDNA.

Electrical Properties of MDCK Cells

1985
The electrical characterization of a transporting epithelium requires information three main aspects: (1) the overall electrical properties across the whole cell layer; (2) the transcellular permeation route; and (3) the paracellular pathway. In the present article we describe assumptions, techniques, efforts, and information collected on these aspects

Endocytosis in MDCK Cells

1992
In polarized cells the plasma membrane is divided into apical and basolateral domains. Membrane is internalized from both these domains and the internalized components can follow several different routes, for example, they can be transported to the opposite domain of the cell (transcytosis), routed to lysosomes or recycled back to the same domain ...

SNARE Protein Trafficking in Polarized MDCK Cells

Traffic, 2000
A key feature of polarized epithelial cells is the ability to maintain the specific biochemical composition of the apical and basolateral plasma membrane domains. This polarity is generated and maintained by the continuous sorting of apical and basolateral components in the secretory and endocytic pathways.

Mineralocorticoid-induced membrane proteins in MDCK cells

Molecular and Cellular Endocrinology, 1982
The Madin-Darby canine kidney (MDCK) cell line, which exhibits properties indicative of a distal tubule origin, evidently binds and responds to mineralocorticoid hormones. We investigated the effects of aldosterone and deoxycorticosterone on protein synthesis in MDCK cells grown either in medium supplemented with serum or in a hormonally defined, serum-
