One of the major defense mechanisms against the development of cancer is governed by the tumor-suppressor protein p53. The p53 protein can induce growth arrest and apoptosis, and, owing to these important functions in controlling cell proliferation, p53 itself has to be tightly regulated.

The murine double minute 2 (mdm2) gene encodes a negative regulator of the p53 tumor suppressor. Amplification of mdm2 or increased expression by unknown mechanisms occurs in many tumors. Thus, increased levels of MDM2 would inactivate the apoptotic and cell cycle arrest functions of p53, as do deletion or mutation of p53, common events in the genesis ...

Interaction between p53 and the E3 ubiquitin ligase murine double minute 2 homolog (MDM2) is regarded as an important target for anticancer therapeutics. The p53/MDM2 protein-protein interaction is one of the best-studied and most targeted intracellular processes related to cancer progression.
