The Role of the Melanocortin‐3 Receptor in Cachexia
Annals of the New York Academy of Sciences, 2003Abstract: Cachexia refers to a synergistic combination of a dramatic decrease in appetite and an increase in metabolism of fat and lean body mass. This combination is found in a number of chronic diseases and is an important determinant of mortality. In this paper, we provide evidence that in both acute and chronic disease models, blockade of the MC4 ...

Melanocortin-3 receptor activates MAP kinase via PI3 kinase
Regulatory Peptides, 2007HEK 293 cells stably expressing human melanocortin-3 receptor (MC3R) were exposed to melanocortin receptor agonist, NDP-MSH (10(-)(10)-10(-)(6) M). ERK1/2 was phosphorylated in a dose-dependent manner with an EC(50) of 3.3+/-1.5 x 10(-)(9) M, similar to the IC(50) of NDP-MSH binding to the MC3R.

Functional characterization of the mouse melanocortin 3 receptor gene promoter
Gene, 2015Melanocortin receptor 3 (MC3R) is expressed in the hypothalamus and pituitary in humans and rodents, and is involved in the control of feeding, energy metabolism, and pituitary function. In the mouse pituitary, MC3R is detected in mammotrophs. This study aimed to clarify the regulatory mechanism for Mc3r expression in the mouse pituitary.

Pharmacology of the giant panda (Ailuropoda melanoleuca) melanocortin-3 receptor
General and Comparative Endocrinology, 2019The melanocortin-3 receptor (MC3R) is a member of the G protein-coupled receptor superfamily that plays a critical role in controlling energy balance and metabolism. Although pharmacological characterization of MC3R has been reported previously in several other species, there is no report on the MC3R from giant panda (Ailuropoda melanoleuca).

Melanocortin 3 receptor (MC3R) gene variants in extremely obese women
International Journal of Obesity, 2000Following several reports of linkage of obesity related phenotypes to human chromosome 20q we sought to determine whether variations of the melanocortin 3 receptor (MC3R) gene are associated with obesity. We screened the MC3R gene coding region and approximately 2 kb of 5' and 3' flanking sequences for DNA variants in unrelated extremely obese women and

Mutations in Melanocortin-3 Receptor Gene and Human Obesity
2016The prevalence of obesity calls for novel therapeutic targets. The melanocortin-3 receptor (MC3R) has been increasingly recognized as an important regulator of energy homeostasis and MC3R has been intensively analyzed in molecular genetic studies for obesity-related traits.

Activation and Endocytic Internalization of Melanocortin 3 Receptor in Neuronal Cells
Annals of the New York Academy of Sciences, 2007Abstract: Melanocortins play a central role in autonomic modulation of metabolism by acting through a family of highly homologous G protein–coupled receptors. Studies with gene knockout mice have implicated neural melanocortin receptors, MC3R and MC4R, in the etiology of obesity, insulin resistance, and salt‐sensitive hypertension.

Postnatal Expression of Melanocortin-3 Receptor in Rat Diencephalon and Mesencephalon
Neuropharmacology, 1997In situ hybridization was applied to examine the postnatal expression of melanocortin-3 (MC-3) receptor mRNA in the rat brain. Very weak and limited signals were seen in the hypothalamus on postnatal day 0 (P0) and in the dorsal lateral thalamus on P4. A marked increase was noted in several regions of the diencephalon and mesencephalon on P7.

This chapter deals with perhaps the least understood receptor for the melanocortin peptides, that is the melanocortin-3 receptor (MC3-R). Although naturally occurring and genetically engineered mutations have provided us with insight into the function of the other known melanocortin receptors, little is known about the physiologic role of the MC3-R ...

Peripheral Melanocortin 3 Receptor (MC3R) Regulates Hepatic Autophagy in Obesity
Diabetes, 2018We have previously reported homozygosity for two MC3R SNPs (C17A+G241A) that partially inactive MC3R increases adiposity in humans and mouse models. However, the precise mechanisms by which hypoactive MC3R alters metabolic homeostasis remain unclear. Given the essential role of hepatic autophagy in cellular mechanisms of obesity, we hypothesized that ...
