Mitochondrial Localization of Cx26 Promotes Colorectal Cancer Malignancy and Exposes a Fatal Vulnerability to Drug Targeting

open access: yesAdvanced Science, EarlyView.
Mitochondrial localization of Cx26 emerges as a double‐edged sword in colorectal cancer. PIMT‐mediated methylation of Cx26 promotes its mitochondrial localization and stability. Mitochondrial Cx26 then facilitates ER‐mitochondria communication and Ca2+ signaling, thereby promoting metastatic progression. Bortezomib exploits this malignant adaptation by

scH16S‐Seq Maps Klebsiella pneumoniae‐Associated Myeloid States and Reveals a CD38–NAD Immunometabolic Axis That Impairs Lysosomal Acidification in Sepsis

open access: yesAdvanced Science, EarlyView.
Single‐cell host–bacterial 16S co‐sequencing (scH16S‐seq) integrates cell‐associated bacterial 16S‐derived signals with host transcriptomes to resolve bacterial signal‐enriched myeloid states in Klebsiella pneumoniae sepsis. Complementary pharmacological and genetic analyses define a CD38–NAD immunometabolic axis connecting mitochondrial bioenergetic ...

Cell Segmentation Beyond 2D—A Review of the State‐of‐the‐Art

open access: yesAdvanced Intelligent Discovery, EarlyView.
Cell segmentation underpins many biological image analysis tasks, yet most deep learning methods remain limited to 2D despite the inherently 3D nature of cellular processes. This review surveys segmentation approaches beyond 2D, comparing 2.5D and fully 3D methods, analyzing 31 models and 32 volumetric datasets, and introducing a unified reference ...

L‐Cysteine and N‐Acetylcysteine Supplementation Improves Clinical Outcome in a Patient With COXPD10

open access: yesAmerican Journal of Medical Genetics Part A, EarlyView.
ABSTRACT MTO1 is a nuclear gene that encodes a mitochondrial protein essential for modifying mitochondrial transfer RNAs (tRNAs) and stabilizing codon‐anticodon interactions to ensure accurate and efficient mitochondrial protein synthesis and oxidative phosphorylation.

SDPR–STK38 axis controls the proliferation–differentiation balance in alveolar type II cells

open access: yesAnimal Models and Experimental Medicine, EarlyView.
The present study identifies SDPR as a pivotal regulator orchestrating the balance between proliferation and differentiation in alveolar type II (AT2) cells. In SDPR+/+ cells, SDPR binds to and inhibits STK38 activity, thereby sustaining GSK‐3β signaling functionality to promote cyclin D1 degradation and maintain cell cycle homeostasis.

Site‐Specific Protein Bioconjugation Through Cellular Incorporation of Noncanonical Amino Acids

open access: yesAngewandte Chemie, EarlyView.
Genetic code expansion (GCE) enables site‐specific installation of noncanonical amino acids containing bioorthogonal conjugation handles, allowing precise, homogeneous protein modification. This review examines the principles of orthogonal translation, surveys the chemistries available for chemoselective labeling, and highlights emerging multi‐site ...
