Esketamine and ketamine are widely used for perioperative analgesia and anesthesia. Despite their established roles in analgesia, sedation, and anesthesia, as well as emerging antidepressant, anti‐tumor, and anti‐inflammatory effects, their clinical use is limited due to side effects and addiction potential.

In Parkinson's disease, SHMT1 downregulation disrupts its interaction with PEMT in astrocytes, reducing SAM levels. This leads to H3K4me1 hypomethylation and decreased Slc1a2/Glul expression, ultimately exacerbating neuroexcitotoxicity and dopaminergic neuron loss.

In Parkinson's disease, the death of dopamine neurons in the substantia nigra, resulting in a decrease in dopamine levels in the striatum. This leads to striatal damage through the HMGB1/RAGE/NF‐κB signaling pathway, thereby inducing motor and cognitive dysfunction.
