Hydrolysis of methylprednisolone hemisuccinate to methylprednisolone by a nonspecific carboxylesterase

Summary Enzymatic hydrolysis of methylprednisolone hemisuccinate with a non-specific carboxylesterase from hog liver was examined. The carboxyl-exterase was found to be useful in hydrolyzing methylprednisolone hemisuccinate to methylprednisolone.

Kinetics of methylprednisolone and its hemisuccinate ester

Methylprednisolone in the form of its hemisuccinate ester was injected intravenously in doses of 10 mg/kg and 63.1 mg. Plasma levels of methylprednisolone and of the ester were measured and their kinetics were calculated. Results indicate dose dependency in the kinetics of both. About 10% of the dose was excreted unchanged as hemisuccinate in the urine,

Initial Rate Studies of Hydrolysis and Acyl Migration in Methylprednisolone 21-Hemisuccinate and 17-Hemisuccinate

The degradation of methylprednisolone 21-succinate in aqueous solution was examined as a function of pH at 25 degrees by monitoring the initial rates of product formation. In addition to hydrolysis, acyl migration fro the 21-hydroxyl group was found to be an important reaction.
