t(4;11) leukemias display addiction to MLL-AF4 but not to AF4-MLL [PDF]

open access: yesLeukemia Research, 2011
The most frequent MLL-gene rearrangement found in leukemia is a reciprocal translocation with AF4 on chromosome 4 resulting in the formation of the MLL-AF4 and the AF4-MLL fusion genes. The oncogenic role of MLL-AF4 is documented but the significance of the reciprocal product - AF4-MLL in leukemia is less clear.

A human genome editing-based MLL::AF4 B-cell ALL model recapitulates key cellular and molecular leukemogenic features.

Blood, 2023
The cellular ontogeny and location of the MLL-breakpoint influence the capacity of MLL-edited CD34+ HSPCs to initiate pro-B-ALL, and recapitulate the molecular features of MLL-AF4+ infant B-ALL patients.

Gene Expression Network and Circ_0008012 Promote Progression in MLL/AF4 Positive Acute Lymphoblastic Leukemia.

Recent Patents on Anti-Cancer Drug Discovery, 2022
BACKGROUND Acute lymphoblastic leukemia with MLL/AF4 rearrangement remains a major hurdle to improving outcomes. Gene network and circRNAs have been found to participate in tumorigenesis, while their roles in leukemia still need to be explored.

Methylation-mediated repression of microRNA-143 enhances MLL–AF4 oncogene expression [PDF]

open access: yesOncogene, 2011
Fusion proteins containing the amino terminus of mixed lineage leukemia (MLL) are common in acute lymphoblastic leukemia (ALL) due to translocations. The MLL-AF4 fusion protein is generated by the translocation t(4;11)(q21;q23), and t(4;11)-positive ALL patients (MLL-AF4 ALL), have a notoriously poorer prognosis compared with patients with other MLL ...

H3K79 Methylation Profiles Define Murine and Human MLL-AF4 Leukemias [PDF]

open access: yesCancer Cell, 2008
We created a mouse model wherein conditional expression of an Mll-AF4 fusion oncogene induces B precursor acute lymphoblastic (ALL) or acute myeloid leukemias (AML). Gene expression profile analysis of the ALL cells demonstrated significant overlap with human MLL-rearranged ALL.
