Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Agile Development of PHP Websites: A Model-Aware Approach

open access: yesComplex Systems Informatics and Modeling Quarterly, 2020
The Design Science research method was hereby employed to develop an artifact that demonstrates the experimental “model-aware” software engineering methodology in the context of PHP Web development – a “low code” development approach with code templates ...

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Compositionality and Refinement in Model-Driven Engineering [PDF]

open access: yes, 2012
Model-driven engineering involves the automatic generation of software artifacts from models of structure and functionality. The use of models as 'source code' has implications for the notions of composition and refinement employed in the modelling language. This paper explores those implications in the context of object-oriented design: establishing a

Mapping customer needs to engineering characteristics: an aerospace perspective for conceptual design

open access: yes, 2014
Designing complex engineering systems, such as an aircraft or an aero-engine, is immensely challenging. Formal Systems Engineering (SE) practices are widely used in the aerospace industry throughout the overall design process to minimise the overall ...

Quality Issues on Model-Driven Web Engineering Methodologies [PDF]

open access: yes, 2009
Nowadays, there are several development methodologies in the field of model-driven web engineering (MDWE) which involve different levels of modeldriven architecture (MDA): CIM, PIM, PSM, or code. Attending to the high number of available methodologies,

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

A methionine‐lined active site governs carbocation stabilization and product specificity in a bacterial terpene synthase

open access: yesFEBS Letters, EarlyView.
This study reveals a unique active site enriched in methionine residues and demonstrates that these residues play a critical role by stabilizing carbocation intermediates through novel sulfur–cation interactions. Structure‐guided mutagenesis further revealed variants with significantly altered product profiles, enhancing pseudopterosin formation. These

Semantic model driven engineering

open access: yes, 2020
Semantic interoperability is a concern in the design and implementation of modern information systems. Whenever two systems communicate it is important that they share an adequate, common interpretation of any information transferred. Similarly, whenever data created or ac- quired in one context is used in another, it is important that the provenance ...
