Shared pathway-specific network mechanisms of dopamine and deep brain stimulation for the treatment of Parkinson’s disease

open access: yesNature Communications
Deep brain stimulation is a brain circuit intervention that can modulate distinct neural pathways for the alleviation of neurological symptoms in patients with brain disorders.

Cognitive and affective Theory of Mind abilities in Parkinson’s disease before and 1 year after subthalamic deep brain stimulation

open access: yesFrontiers in Human Neuroscience
BackgroundParkinson’s disease (PD) is associated with specific cognitive deficits and dysfunctions in Theory of Mind (ToM), which can impact patients’ quality of life. ToM refers to the ability to infer the mental and emotional state of one’s counterpart

KDAC6 alters cell morphology and motility through positive and negative modulation of F‐actin distribution

open access: yesFEBS Open Bio, EarlyView.
KDAC6 has been associated with cell motility and actin structures, but specific domain contributions are not established. The two catalytic domains have differential effects on motility and F‐actin regulation, affecting cortical F‐actin density, motility, stress fibers, and cell spreading.

Evolution‐guided yeast complementation reveals functional differences in human PSPH variants

open access: yesFEBS Open Bio, EarlyView.
Ancient genomes can help guide which human genetic variants are tested experimentally. This study applies that idea to PSPH, a gene involved in serine biosynthesis, and uses high‐throughput yeast complementation to compare variant function. The findings reveal measurable differences among selected alleles and illustrate the value of evolution‐guided ...

Decoding the impact of visual states on adaptive deep brain stimulation feedback signals in movement disorders

open access: yesnpj Parkinson's Disease
We investigated the impact of visual states on basal ganglia oscillatory biomarkers, comparing local field potentials (LFPs) dynamics between Parkinson’s disease (PD) and dystonia and developing a decoding model for state identification.

Heterotropic regulation and negative homotropic cooperativity

open access: yesFEBS Open Bio, EarlyView.
We identified a structural module common to some proteins that couple negative cooperativity with heterotropic regulation, two features that rarely coexist. These proteins are ring‐like and present an ordered asymmetry whereby noncontacting subunits are symmetric, and their tertiary structure differs from that of contacting subunits.

From patient advocacy to patient‐driven research: Building active partnerships beginning at the bench to reach the bedside

open access: yesFEBS Open Bio, EarlyView.
Research is strongest when conducted alongside patients, not just about them. Patient research organizations help integrate patient perspectives into research priorities, study design, and scientific meetings, leading to meaningful patient outcomes and development of relevant therapies.

Multiple system atrophy: Diagnostic challenges and a proposed diagnostic algorithm

open access: yesClinical Parkinsonism & Related Disorders
Multiple system atrophy (MSA) is a heterogenous condition, presenting with core clinical features of autonomic dysfunction, parkinsonism, and/or cerebellar ataxia. The presence of alpha-synuclein glial cytoplasmic inclusion is the hallmark of MSA.

The C‐terminal domain of yeast Arginyltransferase1 is essential for its catalytic activity

open access: yesFEBS Open Bio, EarlyView.
Arginyltransferase 1 (Ate1), a eukaryotic enzyme, catalyses arginylation, transferring arginine from tRNA‐Arg to the amino terminus of the target protein. Overexpression of Ate1 in yeast is lethal and is dependent on arginylation. This study elucidates how mutations in the cofactor‐binding and active site of Ate1 and truncation of its structural ...
