Ultrahigh Speed Direct PCR: A Method for Obtaining Y-STR and STR Based Genotypes in Under 20 Minutes

open access: yes, 2018
There are many situations in forensic DNA typing where the time it takes to identify an individual is critical. Examples include suspects under arrest and individuals detained at ports of entry.

Gut Mycobiota‐Associated Tryptophan Catabolites Protect Against Metabolic Dysfunction‐Associated Steatotic Liver Disease

open access: yesAdvanced Science, EarlyView.
ABSTRACT Accumulating evidence suggests that the intestinal microbiota participates in the progression of metabolic dysfunction‐associated steatotic liver disease (MASLD) through microbiota‐host interaction. However, the beneficial role of commensal mycobiota in MASLD progression remains poorly understood.

Genome‐Wide CRISPR Screen Identifies a microRNA Orchestrating Pleiotropic Resistance to Targeted Therapy and T Cell Immunity in Melanoma

open access: yesAdvanced Science, EarlyView.
A genome‐wide microRNA CRISPR screen identifies miR‐18a as a master regulator of cross‐resistance in melanoma. Loss of miR‐18a activates the AJUBA–YAP/Hippo axis to confer BRAFi resistance and enhances THBS1–CD47 interaction to impair CD8+ T cell immunity. hnRNP A1 is identified as an upstream regulator of miR‐18a processing.

Evaluation of direct PCR for forensic DNA profiling and the development of a direct PCR multiplex

open access: yes, 2012
The use of direct PCR with different types of sample was explored in this study. Genomic DNA preparations at various concentrations and buccal cell counts were deposited on commonly encountered substrates, recovered and amplified using direct PCR before ...

PRMT1‐Mediated LDHA Methylation Drives STAT3 Lactylation to Orchestrate Intestinal Inflammation and Tumorigenesis

open access: yesAdvanced Science, EarlyView.
This study identifies an immunometabolic axis wherein SAM‐driven PRMT1 methylates LDHA, enhancing its activity. The resultant lactate induces STAT3 K709 lactylation, which stabilizes an active conformation to promote STAT3 phosphorylation and IL‐10 expression.

ZDHHC9‐Mediated Palmitoylation of ACSL4 Drives Ferroptosis in Diabetes Mellitus–Induced Erectile Dysfunction

open access: yesAdvanced Science, EarlyView.
ABSTRACT Diabetes mellitus‐induced erectile dysfunction (DMED) is a highly prevalent complication among diabetic patients; however, its underlying pathogenic mechanisms remain incompletely understood. Metabolic disorder is a hallmark of diabetes, yet its precise contribution to DMED progression is not well defined.

Screening for circulating RAS/RAF mutations by multiplex digital PCR

open access: yes, 2016
Recent years have shown a large interest in the application of liquid biopsies in cancer management. Circulating tumor DNA (ctDNA) has been investigated for potential use in treatment selection, monitoring of treatment response, and early detection of ...

Activation of ERBB4 Pathway Inhibits Pathological Transdifferentiation of Lung Epithelial Progenitors into CD66c+ Basal Cells in Severe Lung Injury

open access: yesAdvanced Science, EarlyView.
In fibrotic distal lung regions, CD66c+ basal cells emerge as a pathological state. Using human distal lung organoids, this study identifies CD66c+ basal cells as a pro‐fibrotic state arising through transdifferentiation from secretory, AT2, and basal cells.

Time‐Multiplexed Organic Electrochemical Transistor for Saliva‐Based Rapid Detection of Viral Proteins

open access: yesAdvanced Science, EarlyView.
A time‐multiplexed nanobody‐functionalized organic electrochemical transistor enables rapid and sensitive detection of three respiratory viral proteins from saliva. Sequentially addressed multi‐gate electrodes on a shared channel achieve around 1 fm detection within 15 min without sample preprocessing. Clinical validation demonstrates high specificity,
