Transposon Insertional Mutagenesis in Rice
2010Insertion mutants offer one of the direct ways to relate a gene to its function by employing forward or reverse genetics approaches. Both T-DNA and transposon insertional mutants are being produced in several crops, including rice, the first cereal with its complete genome sequenced.

Insertional mutagenesis in mice
Trends in Genetics, 1987Abstract Insertional mutagenesis via the introduction of exogenous DNA into the germ line has been accomplished in many organisms. In addition to causing the mutation, the introduced DNA serves as a tag for molecular cloning of the affected locus. A number of mutations due to the insertion of either retroviral or microinjected cloned DNA have been ...

Efficient insertional mutagenesis in Streptococcus thermophilus
Gene, 2000Bacteria have always been considered ideal organisms for genetic analysis. While this is true for some model organisms, like Escherichia coli, Bacillus subtilis and, more recently, Lactococcus lactis, genetic analysis of other organisms is often prevented by lack of valuable tools, like vectors, transposons and methods for transformation, gene ...

T-DNA insertional mutagenesis in Arabidopsis
Plant Molecular Biology, 1992T-DNA insertion mutagenesis is one of those recently developed genetic techniques which can be expected to have a major impact in plant molecular biology [17, 50, 87]. Unlike conventional reviews, this brief outlook intends to give a practical insight into current problems and future applications of T-DNA tagging by underlining the potential of the ...

Retroviral-Mediated Insertional Mutagenesis in Zebrafish
2004Publisher Summary This chapter discusses retroviral-mediated insertional mutagenesis in zebrafish. Large-scale chemical mutagenesis screens have resulted in the isolation of thousands of mutations in hundreds of genes that affect zebrafish embryonic development.

Saturation Mutagenesis by Codon Cassette Insertion
Cold Spring Harbor Protocols, 2019Saturation mutagenesis by cassette insertion introduces a library of site-specific changes into a specific DNA sequence within a target gene and is especially useful for analyzing the effect of specific residues on the structure and function of a protein.

Tn10 insertional mutagenesis in Pasteurella multocida
Veterinary Microbiology, 1996The objective of this study was to identify a transposon that will randomly and stably integrate in the genome of Pasteurella multocida. Using the suicide conjugative delivery system, pLOF, containing a kanamycin resistance marked Tn10 insertion element, we determined that Tn10 is effective in producing insertional mutations in this species.

Transposon Insertional Mutagenesis Models of Cancer
Cold Spring Harbor Protocols, 2014Transposon-based insertional mutagenesis in the mouse provides a powerful approach for identifying new cancer genes. Transposon insertions in cancer genes are selected during tumor development because of their positive effect on tumor growth, and the transposon insertion sites in tumors thus serve as tags for identifying new cancer genes.

Insertional mutagenesis in Arabidopsis thaliana
Plant Science, 1991Abstract Recently, a number of mutant gene loci in the Arabidopsis thaliana plant genome have been identified through insertional mutagenesis. In this review, we evaluate different methods used for Agrobacterium tumefaciens -mediated T-DNA insertional mutagenesis with regard to their mutation frequencies and conclude that a major breakthrough in ...

Tnt1 Insertional Mutagenesis in Medicago truncatula
2018Legumes play irreplaceable roles in sustainable agriculture due to their unique capability of fixing gaseous nitrogen in the atmosphere and turning into plant-usable ammonium through interaction with rhizobia. With the completion of genome sequencing of several model and non-model legumes, it is highly desirable to generate mutant populations for ...
