Biphasic Oxidation of Mitochondrial NAD(P)H

The redox state of mitochondrial pyridine nucleotides is known to be important for structural integrity of mitochondria. In this work, we observed a biphasic oxidation of endogenous NAD(P)H in rat liver mitochondria induced by tert-butylhydroperoxide. Nearly 85% of mitochondrial NAD(P)H was rapidly oxidized during the first phase.

Reduction by a model of NAD(P)H

Abstract Kinetics for the reduction of 2-acetylpyridine and 2-benzoylpyridine with 1-benzyl-1,4-dihydronicotinamide in acetonitrile has been studied. Isotope effects of the reduction reveal that the transfer of hydrogen nucleus is involved in the rate-determining step.
