Identification and Functional Analysis of the NADK Gene Family in Wheat

NAD(H) kinase (NADK) is a key enzyme for maintaining the cellular NAD(H)/NADP(H) balance and for NADP(H)-based metabolic pathways. We identified four NADK homologs, NADK1–4, from three major cereal crops, wheat, rice and maize, based on their phylogenetic relationships and gene structures.

Palmitoylation-dependent activation of NADK promotes NADP+ synthesis and tumorigenesis

Abstract NAD kinase (NADK) is the sole cytosolic enzyme that catalyzes the synthesis of nicotinamide adenine dinucleotide phosphate (NADP+) from NAD+. NADP+ is essential for anabolic reactions and redox balance. Here, we show that palmitate facilitates NADP+ synthesis by enhancing NADK palmitoylation and activity.

We have examined sequence diversity of 147 co-crystal structures of proteins utilizing NAD as a cofactor or as a substrate. Most of these proteins bind NAD in an extended conformations. However, there are clear exceptions to this general trend. Several bacterial reductases bind NAD in an extremely folded conformations.
