NADK-mediated proline synthesis enhances high-salinity tolerance in the razor clam

Comparative Biochemistry and Physiology Part A, Molecular & Integrative Physiology
Euryhaline organisms can accumulate organic osmolytes to maintain osmotic balance between their internal and external environments. Proline is a pivotal organic small molecule and plays an important role in osmoregulation that enables marine shellfish to tolerate high-salinity conditions. During high-salinity challenge, NAD kinase (NADK) is involved in

NADK upregulation is an essential metabolic adaptation that enables breast cancer metastatic colonization

2022
Abstract Metabolic reprogramming and metabolic plasticity allow cancer cells to fine-tune their metabolism to adapt to the ever-changing environments of the metastatic cascade, for which lipid metabolism and oxidative stress are of particular importance.

NADK is Activated by Oncogenic KRAS Signaling to Sustain Pancreatic Ductal Adenocarcinoma

SSRN Electronic Journal, 2020
Metabolic adaptations and the signaling events that control them have been implicated in the adaptation of pancreatic ductal adenocarcinoma (PDAC) to survive stresses associated with nutrient and oxygen deprivation at the tumor site. Recently, rewiring of NADPH production has been shown to play a key role in this process.

Schisandra total lignans ameliorate neuronal ferroptosis in 3xTg-AD mice via regulating NADK/NADPH/GSH pathway

Phytomedicine
Alzheimer's disease (AD) is a progressive neurodegenerative disorder with limited treatments. Schisandra total lignans (STL), the primary active component of Schisandra chinensis, shows potential in alleviating AD-related symptoms, though the mechanisms remain unclear.Considering the promoting effect of neuronal ferroptosis on AD and the ...

Investigation of the Relationship Between NADK and RSL3 and Exploration of Potential Usage of RSL3: Potential Applications of RSL3: Insights from Its Relationship with NADK

Theoretical and Natural Science
Lung cancer remains the leading cause of cancer-related deaths, and ferroptosis has emerged as a potential therapeutic target due to its role in tumor suppression. RSL3, a GPX4 inhibitor, and NADK knockdown both promote ferroptosis, but their relationship in lung cancer treatment remains unexplored.

NUAK1-Mediated Phosphorylation of NADK Mitigates ROS Accumulation to Promote Osimertinib Resistance in Non–Small Cell Lung Carcinoma

Cancer Research
Abstract Osimertinib, a third generation epidermal growth factor receptor tyrosine kinase inhibitor, is approved as a first-line therapy in patients with advanced non–small cell lung carcinoma (NSCLC) with EGFR-activating mutations or the T790M resistance mutation.

Abstract 1800: Cytosolic NADK is conditionally essential for folate-dependent nucleotide synthesis in human cells

Cancer Research
Abstract The genes that are required for the growth of mammalian cells can depend on an interplay of cell-intrinsic factors and environmental context. However, there is often little investigation into how nutrient availability impacts gene essentiality. Moreover, efforts such as DepMap have catalogued genes that contribute to cancer cell

Palmitoylation-dependent activation of NADK promotes NADP + synthesis and tumorigenesis

Abstract NAD kinase (NADK) is the sole cytosolic enzyme that catalyzes the synthesis of nicotinamide adenine dinucleotide phosphate (NADP + ) from NAD + .

Abstract 7244: AMPK interacts with PRC1.1 complex on chromatin to regulate NADK expression in response to metabolic stress in acute lymphoblastic leukemia

Cancer Research
Abstract Acute lymphoblastic leukemia (ALL) remains a leading cause of pediatric cancer-related mortality. ALL cells are particularly vulnerable to metabolic stress following activation of AMP-activated protein kinase (AMPK), a master regulator of cellular energy homeostasis.

Selective inhibition of nicotinamide adenine kinases (NADKs) by compact NAD mimics

2009
We have examined sequence diversity of 147 co-crystal structures of proteins utilizing NAD as a cofactor or as a substrate. Most of these proteins bind NAD in an extended conformations. However, there are clear exceptions to this general trend. Several bacterial reductases bind NAD in an extremely folded conformations.
