Pharmacological characterization of nalorphine, a kappa 3 analgesic.

Nalorphine is an unusual opiate. Whereas low doses of nalorphine antagonize morphine analgesia, higher nalorphine doses are analgesic, with ED50 values (95% CL) of 13.4 (11.5, 15.8) mg/kg in the writhing and 39.5 (26.6, 60.1) mg/kg in the tail-flick assay.

Abstract It has been shown that nalorphine changes the LD 50 of the intraperitoneally administered dextropropoxyphene from 68 mg/kg to 105 mg/kg. Nalorphine 5 mg im caused a significant increase in serum PRL in 8 healthy volunteers. Pretreatment with bromocriptine 5 mg inhibited the nalorphine-induced release of PRL, whereas sulpiride administration enhanced it.
