Mutant NPM1 in Acute Myeloid Leukemia Initiation and Maintenance
NPM1 mutations drive acute myeloid leukemia by acting as neomorphic transcriptional regulators that cooperate with Menin–MLL and XPO1 to sustain HOX/MEIS1 expression and block differentiation. Targeting these mutant‐specific transcriptional dependencies provides a rational therapeutic strategy for NPM1‐mutated AML.

ABSTRACT Introduction Spinal cord infarction (SCI) is a rare but devastating myelopathy, characterized by a high disability rate and an unfavorable prognosis. It has often been underdiagnosed and misdiagnosed as idiopathic transverse myelitis (ITM). This study aimed to describe the clinical features, radiological biomarkers, treatments, and functional ...

Paracrine signaling in stem cell renewal and in neoplastic tumor growth [PDF]
Paracrine pathway activities are being increasingly recognized as instrumental regulatory mechanisms of epithelial-stromal interactions that play important roles in physiological and pathological self-renewal of stem cells and in the initiation and maintenance of neoplastic tumor development.

Microengineered Gradient Hydrogels for Mechanobiology
Gradient hydrogels are used to mimic the mechanical heterogeneity in native tissues, offering powerful in vitro platforms to study cell‐material interactions in diverse pathophysiological contexts. Here, we present a comprehensive review of the design and experimental considerations for stiffness gradient hydrogels, discussing exemplary achievements ...

Causal Prediction of TP53 Variant Pathogenicity Using a Perturbation‐Informed Protein Language Model
A TP53‐specific predictor, CaVepP53, is developed by fine‐tuning ESMC on experimentally validated variants, quantifying pathogenicity via Euclidean distances. It outperforms general‐purpose models and extends to five cancer genes, enabling interpretable variant classification for precision medicine.

Hypothesis: Are Neoplastic Macrophages/Microglia Present in Glioblastoma Multiforme?
Most malignant brain tumours contain various numbers of cells with characteristics of activated or dysmorphic macrophages/microglia. These cells are generally considered part of the tumour stroma and are often described as TAM (tumour-associated ...

Hypothesis of the basic biological sense of cancer revisited: a putative explanation of Peto's paradox [PDF]
The conventional interpretation of cancer, summarized in the unified genetic theory of carcinogenesis, assumes that the malignant cell is the anatomical and physiological unit of cancer.

β‐Adrenergic Signaling Promotes Anti‐Tumor Immunity in TP53‐mutant Oral Squamous Cell Carcinoma
β‐adrenergic stimulation enhances anti‐tumor immunity in TP53‐deficient oral squamous cell carcinoma by inducing tumor‐derived secretion of CXCL10, which attracts and activates cytotoxic CD8+ T cells. The findings demonstrate that β‐adrenergic signaling alters tumor–immune interactions via CXCL10‐mediated paracrine activation, revealing a neuro‐immune ...

CD47 expression is decreased in hematopoietic progenitor cells in patients with myelofibrosis
Myelofibrosis (MF) is characterized by increased circulating hematopoietic progenitor cells (HPCs), abnormal cytokine levels, and the survival advantage of neoplastic progenitors over their normal counterparts, which leads to progressive disappearance of
