A new branch of mammalian vitamin B6 metabolism: AKR1C‐mediated conversion of pyridoxal to pyridoxine and 4‐pyridoxolactone

open access: yesThe FEBS Journal, EarlyView.
Pyridoxal 5′‐phosphate (PLP) homeostasis relies on salvage enzymes, yet key metabolic branches remain undefined. We identify AKR1C isozymes as previously undescribed contributors that convert pyridoxal into pyridoxine or 4‐pyridoxolactone through reductase and dehydrogenase activities.

Prebiotic aqueous reactions catalyzed by native nickel without hydrogen

open access: yesThe FEBS Journal, EarlyView.
Serpentinizing (H2‐producing) hydrothermal vents are candidate environments for metabolic origin. They generate highly reducing conditions that convert CO2 to formate and methane in abiotic reactions resembling reactions of the acetyl‐CoA pathway of CO2 fixation. They also contain natural catalysts. Native nickel (Ni0), like Fe0, Co0, and their alloys,

Phenotypic characterization of Arabidopsis thaliana mutants that alleviate root growth defects in a vitamin B6 biosynthesis mutant

open access: yesThe FEBS Journal, EarlyView.
Vitamin B6 is a critical molecule for plant metabolism and development. Rsr4‐1, a loss‐of‐function mutant in a vitamin B6 biosynthesis gene, has a distinct short root phenotype. Here we describe five novel suppressor mutants with an rsr4‐1 background that have normalized root growth and distinct phenotypic and molecular characteristics, representing a ...

Blocking protein quality control degradation leads to structural stabilization of DHFR indel variants

open access: yesThe FEBS Journal, EarlyView.
Blocking protein quality control degradation by deleting the E3 ligases Ubr1 or San1 stabilizes many DHFR insertion and deletion variants that are otherwise partially unfolded. These hypomorphic variants regain folding at lower temperatures or upon methotrexate binding, revealing that PQC degradation can remove proteins that are still functionally ...

Physiological media and oxygen levels modulate cancer hallmarks in cultured breast cancer cells

open access: yesThe FEBS Journal, EarlyView.
MCF7 breast cancer cells were cultured in standard conditions (18% O2 and DMEM) and physiological conditions (5% O2 and Plasmax, a medium modeled after the human plasma metabolome). A combined transcriptomic and proteomic approach was used to identify gene expression signatures at the mRNA and protein levels upstream of functional enrichment analyses ...
