Comparison of the azoreductase and nitroreductase from Clostridium perfringens [PDF]

open access: yesApplied and Environmental Microbiology, 1993
The purified azoreductase and nitroreductase of Clostridium perfringens, which have similar electrophoretic properties, both reacted in a Western blot (immunoblot) with a polyclonal antibody raised against the azoreductase. The activity of both enzymes was enhanced by flavin adenine dinucleotide and was inhibited by menadione, o-iodosobenzoic acid, and

Accessing active fragments for drug discovery utilising nitroreductase biocatalysis

open access: yes
Biocatalysis has played a limited role in the early stages of drug discovery. This is often attributed to the limited substrate scope of enzymes not affording access to vast areas of novel chemical space.

tBid‐Mediated Genetic Ablation of Connective Tissue Cells Reveals Their Key Regulatory Function During Limb Regeneration in Axolotls

open access: yesAdvanced Science, EarlyView.
We establish a tBid‐mediated cell ablation system in axolotls, achieve rapid and efficient ablation of multiple cell types, including muscle stem cell, spinal cord cell, and connective tissue (CT) cells. We investigate the role of CT using tBid‐mediated CT ablation and identify its essential role for limb development and regeneration.

Nitroreductase activity of heart lipoamide dehydrogenase [PDF]

open access: yesBiochemical Journal, 1987
A novel reaction catalysed by lipoamide dehydrogenase is described. In the presence of NADH, lipoamide dehydrogenase reduces the nitro group of 4-nitropyridine and 4-nitropyridine N-oxide. The elution profiles from a DEAE-cellulose column for the dehydrogenase and nitroreductase activities are identical.

Pathogen‐Centric Activation of an Azoreductase‐Responsive Antibody‐Antibiotic Conjugate for the Targeted Eradication of MRSA

open access: yesAdvanced Science, EarlyView.
A pathogen‐centric antibody‐antibiotic conjugate (AZO‐AAC) releases antibiotics via bacterial azoreductase, independent of host‐lysosomal function. This mechanism achieves nanomolar eradication of both planktonic and intracellular methicillin‐resistant Staphylococcus aureus (MRSA).

Design, Synthesis, and Evaluation of Doxifluridine Derivatives as Nitroreductase-Responsive Anticancer Prodrugs

open access: yesMolecules
Antimetabolite antitumor drugs interfere with nucleic acid and DNA synthesis, causing cancer cell death. However, they also affect rapidly dividing normal cells and cause serious side effects.

Therapeutic Applications of Stimuli‐Based Release and Engineering of Extracellular Vesicles

open access: yesAdvanced NanoBiomed Research, EarlyView.
This review summarizes the effects of endogenous and exogenous stimuli, their effects on the natural release of extracellular vesicles, as well as their uptake and release. It also gives an overview of stimuli‐responsive EVs and their therapeutic applications. Extracellular vesicles (EVs), nano‐ to microsized lipid bilayer membrane‐bound particles, are

Cell-Specific Chemical Delivery Using a Selective Nitroreductase–Nitroaryl Pair

open access: yes, 2018
The utility of small molecules to probe or perturb biological systems is limited by the lack of cell-specificity. ‘Masking’ the activity of small molecules using a general chemical modification and ‘unmasking’ it only within target cells could overcome ...

Nitroreduction of flutamide by Cunninghamella elegans NADPH: Cytochrome P450 reductase

open access: yesBiochemistry and Biophysics Reports, 2022
The microbial model of mammalian drug metabolism, Cunninghamella elegans, has three cytochrome P450 reductase genes in its genome: g1631 (CPR_A), g4301 (CPR_B), and g7609 (CPR_C).

Environmental Identification of Novel Enzymes for Polyurethane and Polyamide Degradation

open access: yesAngewandte Chemie International Edition, EarlyView.
Heteroatom‐containing plastics such as polyurethanes and nylons are produced at scale yet remain largely unrecycled. Biocatalysts offer a promising route to depolymerization under mild conditions. Here, we present a screening platform for the discovery of novel polyurethane‐ and nylon‐degrading enzymes from environmental samples and demonstrate ...
