In silico and in vitro exploration of a tyrosinase for biocatalytic production of catechols

open access: yesFEBS Open Bio, EarlyView.
Tyrosinase from Ralstonia pseudosolanacearum is a promising biocatalyst for producing valuable catechols from monophenol substrates. This tyrosinase is uniquely suited to this due to its high monophenolase : diphenolase ratio. We combined in silico docking and in vivo kinetic characterisation of this tyrosinase with 11 industrially relevant monophenols,

Adenosine triphosphate as a modulator of protein interactions and stability

open access: yesFEBS Open Bio, EarlyView.
ATP is best known as the cell's energy currency, but it also shapes how proteins fold, interact, aggregate and form biomolecular condensates. This review explains the emerging physical principles behind these effects, including weak binding to charged protein regions, magnesium‐dependent behaviour and concentration‐dependent control of protein ...

Protocol for quantifying miRNA trafficking across the endosomal membrane

open access: yesFEBS Open Bio, EarlyView.
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.

Heterotropic regulation and negative homotropic cooperativity

open access: yesFEBS Open Bio, EarlyView.
We identified a structural module common to some proteins that couple negative cooperativity with heterotropic regulation, two features that rarely coexist. These proteins are ring‐like and present an ordered asymmetry whereby noncontacting subunits are symmetric, and their tertiary structure differs from that of contacting subunits.

Avidin is evolutionarily conserved in fish but dispensable for development and resistance against Streptococcus agalactiae in zebrafish

open access: yesFEBS Open Bio, EarlyView.
The presence of biotin‐binding avidin proteins in fish and their biological significance are poorly characterized. We cataloged fish avidins and demonstrate that they are widely present and evolutionarily conserved. We created avd knockout zebrafish and show that zebavidin is dispensable for development and that resistance of avd knockout embryos in ...

A novel RNA-binding motif in influenza A virus non-structural protein 1

Nature Structural & Molecular Biology, 1997
The solution NMR structure of the RNA-binding domain from influenza virus non-structural protein 1 exhibits a novel dimeric six-helical protein fold. Distributions of basic residues and conserved salt bridges of dimeric NS1(1-73) suggest that the face containing antiparallel helices 2 and 2' forms a novel arginine-rich nucleic acid binding motif.

Recombinant non-structural 1 (NS1) protein of dengue-2 virus interacts with human STAT3β protein

Virus Research, 2005
A combination of yeast two-hybrid library screening, co-immunoprecipitation and immunofluorescence microscopy demonstrated that dengue-2 virus non-structural 1 (NS1) protein can interact with an N-terminally truncated form of human STAT3beta (DeltaN40-STAT3beta) protein.

Expression, purification, and characterization of recombinant NS-1, the porcine parvovirus non-structural protein

Journal of Virological Methods, 2009
The non-structural protein NS-1 of porcine parvovirus (PPV) could be a useful antigen for differentiating pigs infected with PPV from those vaccinated with inactivated whole virus. NS-1 was expressed in Escherichia coli using the pET-32a (+) vector. The fusion protein, which was expressed at a high level, was similar antigenically to the native NS-1 ...

DNA vaccine against the non-structural 1 protein (NS1) of dengue 2 virus

Vaccine, 2006
Dengue is one of the most important mosquito-borne viral disease causing dengue fever and/or dengue shock syndrome/haemorrhagic fever. In some reports, the non-structural protein 1 (NS1) has been identified as a promising antigen for the development of vaccines against dengue virus (DENV).
