Study of Norethindrone in Contraception

Norethindrone was given to 210 women as a contraceptive, at a dose of 10 mg. per day for 20 days of each menstrual cycle, in order to determine what maximal effectiveness could be achieved in a carefully designed field trial. In a total of 6,232 cycles there were no pregnancies and no rejections of the treatment due to side effects.

Bioavailability of norethindrone in human subjects

A competitive protein binding assay for norethindrone was developed to measure plasma levels in human subjects. The plasma levels were considerably higher in women than in men, especially at low dose levels. The plasma levels were directly related to the dose in men; but greater variations in the plasma levels were observed in women.

Bioavailability of norethindrone and menstrual pattern after insertion of norethindrone acetate implants in rhesus monkeys

Subdermal silastic implants releasing norethindrone acetate were inserted in five rhesus monkeys for a period of seven months. The serum norethindrone (NET) levels were estimated from blood samples collected fortnightly. There was an initial high level of NET followed by fluctuating levels of NET in 4 out of five monkeys.

The contraceptive steroid norethindrone reacts with isoniazid both in vivo and in vitro to give the corresponding hydrazone, which exists as syn and anti (with respect to C-4) isomers. These isomers rapidly interconvert, with the anti form predominating in solution.

Influence of aspirin on the pharma of norethindrone

Abstract The influence of a single dose of aspirin on the pharmacokinetics of the synthetic progestogen norethindrone was studied in rabbits. It was found that neither the 24-hr plasma levels nor the pharmacokinetic parameters of norethindrone following intravenous dosing were significantly altered by aspirin.
