A Portable, Digital Nucleic Acid Amplification Test for Use at the Point of Care
Nucleic acid tests are key tools for the detection and diagnosis of many diseases. In many cases, the amplification of nucleic acids is required to reach a detectable level.

This work presents a CRISPR‐based, ultrasensitive multiplex protein detection array capable of simultaneously analyzing six plasma biomarkers associated with Alzheimer's disease (AD). By integrating antibody‐mediated signal transduction, recombinase polymerase amplification, and spatially encoded CRISPR‐Cas12a, the system achieves detection sensitivity

Hemoglobin's α‐Helix‐to‐β‐Sheet Transition Enables Targeted mRNA Delivery to the Lung
Acidic heating converts hemoglobin into β‐sheet‐rich fibrils with positively charged surfaces. These hemoglobin fibrils electrostatically complex IL‐11 scFv mRNA, selectively bind circulating platelets, and hitchhike to the lung, where they are enriched in fibroblasts.

Programming nucleic acid based molecular machineries for homogeneous bioanalyte detection
Accurate diagnosis can make a life-changing difference to therapeutic options and outcomes, and that is highly dependent on the progress of biomarker discovery as well as the precision of analytical tools available.

This study introduces a biomimetic “nanofusion” platform that integrates the biostability of threose nucleic acids (TNA) with homotypic cell‐membrane cloaking to combat drug‐resistant TNBC. By leveraging a non‐canonical membrane‐fusion pathway for direct cytosolic delivery, the platform bypasses endosomal sequestration. To achieve potent AKT2 silencing

This study unveils a pathogenic axis in oral cancer where NUDT21 suppresses a tumor suppressor network, prominently PTEN, via 3'UTR lengthening. To exploit this transcriptomic vulnerability, we developed a biomimetic Nano‐APA‐editor. By delivering CRISPR/Cas9, we reprogrammed the global 3'UTR landscape, restoring multi‐target tumor suppression and ...

An optimized single‐cell transcriptomic framework profiles over 60 000 cells to map the ovine rumen microbiome, partitioning the ecosystem into seven cross‐species functional clusters. In heat‐resistant hosts, a lineage‐specific metabolic shift in Anaerovibrio lipolyticus toward a highly glycolytic phenotype contributes to a “nutritional sparing ...

The KIF6‐RBP Complex Orchestrates mRNA Transport Required for Sperm Flagellar Assembly
Two homozygous deleterious KIF6 variants are identified in unrelated men with impaired sperm motility. Mouse models and multi‐omics analyses reveal that KIF6 cooperates with the RNA‐binding proteins FMRP and FXR1 to deliver mRNAs essential for sperm flagellar assembly, linking disrupted mRNA transport to reduced abundance of key structural and ...

Whole genome amplification for PGD and PND; molecular and a-CGH diagnosis
Whole genome amplification amplifies the entire genome in a few hours from samples of minimal DNA quantities, even from single cells. This may have many applications, especially in prenatal diagnosis, PGD and PGS. The hypothesis for chapter 3 was: Can
