The Multiple Properties and Functions of Nucleolin

Nucleolin, one of the most abundant proteins of the nucleolus is a fascinating protein with multiple proposed functions within and outside the nucleoli. Since its discovery almost 40 years ago, it was proposed that nucleolin is involved in ribosome biogenesis but until recently the molecular mechanisms of this involvement were not known.

Nucleolin Responds to DNA Damage

Stimuli that damage DNA activate stress-signaling pathways that include the activation of the tumor suppressor p53. Daniely et al. report that ionizing radiation (IR) or the drug camptothecin produce a transient p53-dependent redistribution of nucleolin from nucleoli to the nucleoplasm.

Okadaic acid is a potent inhibitor of protein phosphatase type 1 (PP1) and type 2A and induces apoptosis in human osteoblastic Saos-2 and MG63 cells.

Abstract : The major accomplishment of our studies is the finding that nucleolin stabilizes p53 by inhibiting the p53-antagonist Hdm2. The increase in p53 protein by nucleolin leads to higher expression of p21cip1/waf1 a reduced rate of cellular proliferation and an increase in apoptosis.
