Nuclear Export of mRNA by TAP/NXF1 Requires Two Nucleoporin-Binding Sites but Not p15 [PDF]
Metazoan NXF1/p15 heterodimers promote export of bulk mRNA through nuclear pore complexes (NPC). NXF1 interacts with the NPC via two distinct structural domains, the UBA-like domain and the NTF2-like scaffold, which results from the heterodimerization of the NTF2-like domain of NXF1 with p15.

Metazoan NXF1-p15 heterodimers promote the nuclear export of bulk mRNA across nuclear pore complexes (NPCs). In vitro, NXF1-p15 forms a stable complex with the nucleoporin RanBP2/Nup358, a component of the cytoplasmic filaments of the NPC, suggesting a ...

URH49 exports mRNA by remodeling complex formation and mediating the NXF1-dependent pathway
Biochimica et Biophysica Acta (BBA) - Gene Regulatory Mechanisms, 2020The TREX complex integrates information from nuclear mRNA processing events to ensure the timely export of mRNA to the cytoplasm. In humans, UAP56 and its paralog URH49 form distinct complexes, the TREX complex and the AREX complex, respectively, which cooperatively regulate the expression of a specific set of mRNA species on a genome wide scale.

Structure of the C-terminal FG-nucleoporin binding domain of Tap/NXF1
Nature Structural Biology, 2002The vertebrate Tap protein is a member of the NXF family of shuttling transport receptors for nuclear export of mRNA. Tap has a modular structure, and its most C-terminal domain is important for binding to FG repeat-containing nuclear pore proteins (FG-nucleoporins) and is sufficient to mediate nuclear shuttling.

Allostery and inter-domain dynamics in NXF1: An insight into viral CTE-RNA binding
International Journal of Biological MacromoleculesNucleocytoplasmic export of mRNA is a fundamental process in eukaryotic cells, facilitating the transportation of mRNA transcripts from nucleus to cytoplasm. Central to this pathway is Nuclear Export Factor 1 (NXF1), a key RNA binding protein (RBP) mediating mRNA export through the Nuclear Pore Complex (NPC).

Alternative transcripts expressed by small bristles, the Drosophila melanogaster nxf1 gene
Gene, 2010The tissue-specific accumulation of small bristles (Dm nxf1) transcripts at different developmental stages of Drosophila melanogaster was analyzed by Northern blots and RT PCR. We identified four distinct transcripts: ubiquitous (3.5kb); ovary and early embryo specific (3.3kb); testis specific (1.9kb and 2.8kb) and nervous system specific (5.1kb).

Imaging within single NPCs reveals NXF1’s role in mRNA export on the cytoplasmic side of the pore [PDF]
Translocation of mRNA through the nuclear pore complex (NPC) requires interactions with different NPC regions. To determine the interactions that are crucial for effective mRNA export in living cells, we examined mRNA export within individual pores by applying various types of mRNA export blocks that stalled mRNPs at different stages of transition ...

KRAB Zinc Finger protein Znf684 interacts with Nxf1 to regulate mRNA export
2021Abstract Cys2His2 (C2H2) type zinc finger (ZnF) proteins constitute a large class of proteins that are generally considered to be DNA-binding transcription factors. Using affinity purification followed by mass spectrometry, as well as reciprocal co-immunoprecipitation experiments, we determined that the C2H2-ZnF ...

The transcription and transportation of mRNA are coupled processes; however, the mechanisms linking these processes remain unclear. Additionally, the significance of this connection in cancer drug development is poorly understood. To address these issues, we investigated the role of CDK12 kinase, which regulates RNA transcription through the ...

The role of sbr/Dm nxf1 gene in syncytial development in Drosophila melanogaster
Cell and Tissue Biology, 2015Syncytial development is a property of early embryogenesis and spermatogenesis in Drosophila melanogaster. All elements of syncytium are linked in a common cytoskeletal network, which provides equal conditions and synchronous divisions of each nucleus. The cytoskeleton is necessary for the formation and function of the spindle.
