Blending Observational Methods: Possibilities, Strategies, and Challenges

open access: yes, 2008
Qualitative researchers have used the observational strategies of video recording and participant observation to investigate specific phenomena. Although there have been recent advances in the separate use of these strategies, there is evidence that ...

Observing Neurodiversity, Observing Methodology: Ethnography in Pandemic Times

open access: yesInternational Journal of Qualitative Methods
Ethnographic researchers have long relied upon observation as a powerful means to learn about social relations. This paper discusses research observation that was conducted as a part of an institutional ethnography (IE) investigating how people use the language and ideas of neurodiversity across different settings. While our research protocol initially

Cell‐cycle‐specific lesion evolution rather than inhibition of double‐strand‐break repair underpins cisplatin radiosensitization

open access: yesMolecular Oncology, EarlyView.
We analyze cisplatin–DNA adducts (CDAs) and double‐strand breaks (DSBs) in a cell‐cycle‐dependent manner. We find that CDAs form similarly across all cell cycle phases. DSBs arise only in S‐phase. CDAs might not directly impair DSB repair, but S‐phase DSB lesions evolve in the presence of CDAs and disrupt repair in G2, also causing radiosensitization ...

Interaction of HS1BP3 with cortactin modulates TKS5 localisation, cell secretion and cancer malignancy

open access: yesMolecular Oncology, EarlyView.
Here, we demonstrate that HS1BP3 interacts with Cortactin through a proline‐rich region (PRR3.1) and show that this interaction, and HS1BP3 itself, promote cancer cell proliferation and invasion. Inhibition of this interaction leads to build‐up of TKS5 in multivesicular endosomes and altered secretion of CD63 and CD9, providing an explanation for the ...

A Methodology for Effective Metadata Design in Earth Observation

open access: yes, 2017
The satellite images have become an essential source of information to address and analyze environmental issues quickly, repeatedly and in a reliable way. The increasing number of remotely sensed images are the first impediments for data discovery, access and processing.

ESR1 methylation and ESR1 mutations in circulating tumor cells (CTCs) and paired plasma‐cfDNA of advanced breast cancer patients: A feasibility proof‐of‐concept study

open access: yesMolecular Oncology, EarlyView.
Circulating tumor cells (CTCs) and plasma cell‐free DNA (cfDNA) were analyzed to detect ESR1 mutations and methylation in patients with advanced breast cancer. CTC‐derived DNA showed higher sensitivity for mutation detection and revealed complementary genetic and epigenetic alterations, highlighting the added value of CTC analysis for understanding ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Observational Scaffolding for Learning Analytics: A Methodological Proposal

open access: yes, 2018
Temporal analysis of learning data is attracting the interest of researchers, and a growing body of Learning Analytics (LA) research applies lag sequential analysis. However, lack of methodological frameworks that guide the data gathering and analysis poses multiple conceptual, methodological, analytical and technical challenges. While observation as a
