Bromopyruvate, an analogue of pyruvate, one of the substrates of octopine dehydrogenase, was tested as an inhibitor of the enzyme. Provided both the coenzyme and the second substrate, arginine, were present, bromopyruvate rapidly inactivated the enzyme.

Culture media selective for Agrobacterium were inoculated with dilutions of soil and crown gall tumor suspensions. Colonies on the selective media were replica plated on a medium with octopine or nopaline. More than 500 isolates were recovered, about 10% of which were confirmed as octopine-utilizing, fluorescent pseudomonads.

Essential Histidyl Residues of Octopine Dehydrogenase

Octopine dehydrogenase from muscles of Pecten maximus has been treated by diethylpyrocarbonate at pH 6.0. The enzyme was completely inhibited when 2 histidyl residues were carbethoxylated. In the same conditions rabbit muscle lactate dehydrogenase looses its activity when 12 histidines are modified.
