ORAI1 calcium channel orchestrates skin homeostasis [PDF]

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2013
International audienceTo achieve and maintain skin architecture and homeostasis, keratinocytes must intricately balance growth, differentiation, and polarized motility known to be governed by calcium.

Distinct pharmacological profiles of ORAI1, ORAI2, and ORAI3 channels

open access: yesCell Calcium, 2020
The ubiquitous Ca2+\ua0release-activated Ca2+\ua0(CRAC) channel is crucial to many physiological functions. Both gain and loss of CRAC function is linked to disease.

A New Selective Pharmacological Enhancer of the Orai1 Ca2+ Channel Reveals Roles for Orai1 in Smooth and Skeletal Muscle Functions

open access: yesACS Pharmacology and Translational Science, 2020
Store-operated calcium (Ca2+) entry is an important homeostatic mechanism in cells, whereby the release of Ca2+ from intracellular endoplasmic reticulum stores triggers the activation of a Ca2+ influx pathway.

TRPC1 and ORAI1 channels in colon cancer

Cell Calcium, 2019
Colon cancer cells, like other types of cancer cells, undergo the remodeling of the intracellular Ca2+ homeostasis that contributes to cancer cell hallmarks including enhanced cell proliferation, migration, and survival. Colon cancer cells display enhanced store-operated Ca2+ entry (SOCE) compared with their non-cancer counterparts.

Blue light opens the ORAI1 LOC(K)

Cell Calcium, 2021
The current optogenetic tools to elevate cytoplasmic Ca2+ concentrations either do not have high ion selectivity or activate undesirable signaling pathways. He et al. (2021) generated a light-operated Ca2+ channel by integrating a photosensitive module directly into ORAI1 channels to selectively raise cytoplasmic Ca2+ levels.

Visualization of the Orai1 Homodimer and the Functional Coupling of Orai1-STIM1 by Live-Cell Fluorescence Lifetime Imaging

Microscopy and Microanalysis, 2010
AbstractThe Orai1-STIM1 constructed store-operated Ca2+ channels (SOCs) have been found to exert several essential Ca2+ entry/signaling cascades, e.g., the generation of immune response in T lymphocytes. Although biochemical and novel imaging evidence appear to indicate that Orai1 and STIM1 interact with each other to achieve store-operated Ca2+ entry (

The function of the calcium channel Orai1 in osteoclast development

The FASEB Journal, 2021
Abstract To determine the intrinsic role of Orai1 in osteoclast development, Orai1‐floxed mice were bred with LysMcre mice to delete Orai1 from the myeloid lineage. PCR, in situ labelling and Western analysis showed Orai1 deletion in myeloid‐lineage cells, including osteoclasts, as expected.
