SARS-CoV-2 ORF3a Interacts and Stabilizes HMOX1 Expression and Function

2023
This thesis describes the interaction between SARS-CoV-2 Open Reading Frame 3a (ORF3a) and the Heme Oxygenase 1 (HMOX1). SARS-CoV-2 is a novel coronavirus and the causative agent of COVID-19, the highly contagious infection behind the 2019 COVID-19 pandemic.

Insights into Ion Conduction Mechanisms Through the ORF3a Channel by Computational Modelling

Investigating the selectivity and mechanism of the SARSCoV-2 ORF3a protein channel remains a significant challenge within the scientific community. Speculation surrounds its potential calcium ion selectivity, implicating a significant role in the pathology of COVID-19 by influencing viral replication and the cellular virus response.

SARS-CoV-2 protein ORF3a induces Atg8ylation of lysosomal membranes

AbstractAutophagy Conjugation machinery forms a center piece of autophagy and is essential for sequestration of a broad range of cargo destined for degradation. Apart from its role in canonical autophagy, recent evidence suggests an unconventional role of conjugation machinery.

Myelin basic protein antagonizes the SARS-CoV-2 protein ORF3a-induced autophagy inhibition

Biochimie
Inhibition of autophagy is one of the hallmarks of the SARS-CoV-2 infection. Recently it was reported that SARS-CoV-2 protein ORF3a inhibits fusion of autophagosomes with lysosomes via interaction with VPS39 thus preventing binding of homotypic fusion and protein sorting (HOPS) complex to RAB7 GTPase.

ASC Dynamics Associated with ORF3a Protein Expression in Microvascular Lung Endothelial Cells

Proceedings of the Bulgarian Academy of Sciences
The NLRP3 inflammasome is a key sensor of cellular stress and pathogen-associated signals in the innate immune system. Using confocal fluorescence microscopy in microvascular lung endothelial cells, we examined ASC speck formation following expression of the SARS-CoV-2 ORF3a protein.
