Decision-making with multiple correlated binary outcomes in clinical trials [PDF]

open access: yes, 2019
Clinical trials often evaluate multiple outcome variables to form a comprehensive picture of the effects of a new treatment. The resulting multidimensional insight contributes to clinically relevant and efficient decision-making about treatment superiority.

Insights into PI3K/AKT signaling in B cell development and chronic lymphocytic leukemia

open access: yesFEBS Letters, EarlyView.
This Review explores how the phosphoinositide 3‐kinase and protein kinase B pathway shapes B cell development and drives chronic lymphocytic leukemia, a common blood cancer. It examines how signaling levels affect disease progression, addresses treatment challenges, and introduces novel experimental strategies to improve therapies and patient outcomes.

Diphthamide synthesis is linked to the eEF2‐client chaperone machinery

open access: yesFEBS Letters, EarlyView.
The diphthamide modification of eukaryotic translation elongation factor 2 (eEF2) is important for accurate protein synthesis. We addressed the potential coordination of de novo eEF2 synthesis with simultaneous or subsequent diphthamide modification. Our work reveals that the co‐chaperones Hgh1 and Cpr7, which are known to support folding of nascent ...

On the Actionability of Outcome Prediction [PDF]

open access: yesarXiv, 2023
Predicting future outcomes is a prevalent application of machine learning in social impact domains. Examples range from predicting student success in education to predicting disease risk in healthcare. Practitioners recognize that the ultimate goal is not just to predict but to act effectively.

Functional variation among LPMOs revealed by the inhibitory effects of cyanide and buffer ions

open access: yesFEBS Letters, EarlyView.
This study addresses the inhibition of lytic polysaccharide monooxygenases (LPMOs) by cyanide and explains how and why the magnitude of observed inhibitory effects depends on the way LPMO reactions are setup and on the type of LPMO. Enzymes known as lytic polysaccharide monooxygenases (LPMOs) are mono‐copper polysaccharide‐degrading peroxygenases that ...

Real-World Immunosuppressant Treatment Patterns for Patients with Lupus Nephritis in the United States

open access: yesRheumatology and Therapy, 2023
Introduction Lupus nephritis (LN) treatment aims to control and prevent flares and irreversible kidney damage. Around 30% of patients are unresponsive to treatment; however, real-world LN treatment patterns have not been reported.

Ablation of LRP6 in alpha‐smooth muscle actin‐expressing cells abrogates lung inflammation and fibrosis upon bleomycin‐induced lung injury

open access: yesFEBS Letters, EarlyView.
Low‐density lipoprotein receptor‐related protein 6 (LRP6) is a key receptor for the Wnt antagonist Dickkopf1 (DKK1). DKK1 protein expression is induced in a bleomycin (BLM)‐induced lung injury model. We show that DKK1 induces proinflammatory and profibrotic genes in lung fibroblasts.

A qualitative exploration of the patient experience of erosive and non-erosive hand osteoarthritis

open access: yesJournal of Patient-Reported Outcomes, 2021
Background Many patients with hand osteoarthritis (HOA) experience reduced health-related quality of life. This study sought to better understand the disease and treatment experience of individuals with HOA, explore any differences in experiences between

Insertion of the FeB cofactor in cNORs lacking metal inserting chaperones

open access: yesFEBS Letters, EarlyView.
Nitric oxide reductase is an enzyme found in the bacterial denitrification pathway. The NOR active site contains a non‐heme iron, often, but not always inserted with the assistance of chaperones. Here, we study the insertion of FeB in the subfamily of cNORs lacking chaperones and found a putative channel, conserved in the family, perhaps enabling the ...
