PRELIVE: A Framework for Predicting Lipid Nanoparticles In Vivo Efficacy and Reducing Reliance on Animal Testing

open access: yesAdvanced Functional Materials, EarlyView.
PREdicting LNP In Vivo Efficacy (PRELIVE) framework enables the prediction of lipid nanoparticle (LNPs) organ‐specific delivery through dual modeling approaches. Composition‐based models using formulation parameters and protein corona‐based models using biological fingerprints both achieve high predictive accuracy across multiple organs.

Normal high density lipoprotein inhibits three steps in the formation of mildly oxidized low density lipoprotein: steps 2 and 3

open access: yesJournal of Lipid Research, 2000
Treatment of human artery wall cells with apolipoprotein A-I (apoA-I), but not apoA-II, with an apoA-I peptide mimetic, or with high density lipoprotein (HDL), or paraoxonase, rendered the cells unable to oxidize low density lipoprotein (LDL).

Synthesis of oxidized phospholipids by sn-1 acyltransferase using 2–15-HETE lysophospholipids

open access: yesJournal of Biological Chemistry, 2019
Recently, oxidized phospholipid species have emerged as important signaling lipids in activated immune cells and platelets. The canonical pathway for the synthesis of oxidized phospholipids is through the release of arachidonic acid by cytosolic ...

Nano‐ and Micro‐Sized Solid Materials Used as Antiviral Agents

open access: yesAdvanced Functional Materials, EarlyView.
Due to the rise of viral infections in humans and possible viral outbreaks, the use of nano‐ or micro‐sized materials as antiviral agents is rapidly increasing. This review explores their antiviral properties against RNA and DNA viruses, either as a prevention or a treatment tool, by delving into their mechanisms of action and how to properly assess ...

Enzymatically oxidized phospholipids restore thrombin generation in coagulation factor deficiencies

open access: yesJCI Insight, 2018
Hemostatic defects are treated using coagulation factors; however, clot formation also requires a procoagulant phospholipid (PL) surface.

Shedding Light on the Cellular Uptake Mechanisms of Bioactive Glass Nanoparticles as Controlled Intracellular Delivery Platforms: A Review of the Recent Literature

open access: yesAdvanced Healthcare Materials, EarlyView.
This review summarizes the main uptake pathways of bioactive glass nanoparticles (BGNs) and their intracellular localization, highlighting that BGNs are mainly internalized and entrapped within endosomes/lysosomes. Strategies for controlled intracellular ion release, with implications for targeted modulation of cell behavior, are discussed. The need to

Oxidized phospholipids and lipoprotein-associated phospholipase A2 as important determinants of Lp(a) functionality and pathophysiological role

open access: yesJournal of Biomedical Research, 2018
Lipoprotein(a) [Lp(a)] is composed of a low density lipoprotein (LDL)-like particle to which apolipoprotein (a) [apo(a)] is linked by a single disulfide bridge.

New Insights into Atonic Postpartum Hemorrhage: Animal Model Construction Based on Placental Nanodelivery Systems

open access: yesAdvanced Healthcare Materials, EarlyView.
This study develops a placenta‐targeted nanodelivery system co‐loading HMGB1 protein and the NLRP3 agonist nigericin to establish an animal model of atonic postpartum hemorrhage. The model accurately recapitulates clinical phenotypes, including prolonged labor and uterine contractility dysfunction, while revealing inflammatory activation in placental ...

Strategies to Improve the Lipophilicity of Hydrophilic Macromolecular Drugs

open access: yesAdvanced Healthcare Materials, EarlyView.
Hydrophilic macromolecular drugs can be successfully lipidized by covalent attachment of lipids, by hydrophobic ion pairing with negatively or positively charged surfactants, and by dry or wet reverse micelle formation. Lipophilicity enhancement of hydrophilic macromolecules has several benefits including stability and bioavailability improvement ...

Photosensitizing Lipid Nanoparticles for Ferroptosis‐Enhanced Photodynamic Cancer Therapy via GPX4 Silencing

open access: yesAdvanced Healthcare Materials, EarlyView.
Photosensitizing lipid nanoparticles (PLNPs) are engineered by incorporating cholesterol–PEG–pheophorbide a into MC3‐based LNPs and encapsulating GPX4‐targeting siRNA. Upon light activation, PLNPs generate reactive oxygen species (ROS) while silencing GPX4 to induce ferroptosis.
