Rule-based Modelling and Tunable Resolution [PDF]

open access: yes, 2009
We investigate the use of an extension of rule-based modelling for cellular signalling to create a structured space of model variants. This enables the incremental development of rule sets that start from simple mechanisms and which, by a gradual ...

Cleavage‐Resistant CYLD Protects Against Autoimmune Hepatitis

open access: yesAdvanced Science, EarlyView.
Proteolytic cleavage of the deubiquitinase CYLD emerges as a critical driver of autoimmune hepatitis. TNFα‐induced CYLD loss in macrophages amplifies S100A9‐triggered MAPK activation, leading to excessive chemokine production and hepatic inflammation. Pharmacological inhibition of MEK signaling effectively attenuates experimental disease, highlighting ...

Mechanistic studies of anti-hyperpigmentary compounds: elucidating their inhibitory and regulatory actions. [PDF]

open access: yes, 2014
Searching for depigmenting agents from natural sources has become a new direction in the cosmetic industry as natural products are generally perceived as relatively safer.

Combined Photothermal and mTOR‐Targeted Therapy Overcomes Immune Evasion and Enhances Checkpoint Blockade Efficacy in Metastatic Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
This study reveals that photothermal therapy, while inducing immunogenic cell death in triple‐negative breast cancer, paradoxically activates the oncogenic mTOR pathway to drive immune evasion. To counter this, a smart nanocomposite is engineered to co‐deliver localized hyperthermia and mTOR inhibition.

Structural Mechanism for the Specific Assembly and Activation of the Extracellular Signal Regulated Kinase 5 (ERK5) Module [PDF]

open access: yes, 2013
Mitogen-activated protein kinase (MAPK) activation depends on a linear binding motif found in all MAPK kinases (MKK). In addition, the PB1 (Phox and Bem1) domain of MKK5 is required for extracellular signal regulated kinase 5 (ERK5) activation.

Fermented Lacticaseibacillus Paracasei Cultures Ameliorate Colitis by Modulating Microbiota‐Derived Tryptophan Metabolism and Macrophage Polarization

open access: yesAdvanced Science, EarlyView.
A solid‐state fermented probiotic (PYW) is developed using wheat bran as substrate, containing high viable Lacticaseibacillus paracasei and bioactive metabolites. PYW alleviates intestinal inflammation by gut microbiota composition, enriching indole derivatives, activating aryl hydrocarbon receptor signaling, and regulating macrophage polarization. PYW

Differential Response of Heat-Shock-Induced p38 MAPK and JNK Activity in PC12 Mutant and PC12 Parental Cells for Differentiation and Apoptosis [PDF]

open access: yes, 2010
Among the 3 mitogen-activated protein kinases -- ERK, p38 MAPK and JNK -- JNK has been suggested to participate in apoptosis, whereas p38 MAPK is thought to be part of the differentiation response.

Modulating Integrin and Growth Factor Signaling With Peptides: Strategies to Synergistically Enhance Bone Tissue Regeneration

open access: yesAdvanced Science, EarlyView.
It has been demonstrated that, in the bone extracellular matrix (ECM), integrins and growth factor receptors (GFRs) engage in synergistic signaling to guide bone healing and regeneration. This review provides a comprehensive overview of current strategies using ECM‐derived peptides to recreate the cellular microenvironment and harness synergistic ...

The Protective Effect of Beraprost Sodium on Diabetic Nephropathy by Inhibiting Inflammation and p38 MAPK Signaling Pathway in High-Fat Diet/Streptozotocin-Induced Diabetic Rats

open access: yesInternational Journal of Endocrinology, 2016
Background. p38 mitogen-activated protein kinase (MAPK) plays a crucial role in regulating signaling pathways implicated in inflammatory processes leading to diabetic nephropathy (DN).

Beyond antioxidants: the cellular and molecular interactions of flavonoids and how these underpin their actions on the brain [PDF]

open access: yes, 2010
The consumption of flavonoid-rich foods and beverages has been suggested to limit the neurodegeneration associated with a variety of neurological disorders and to prevent or reverse normal or abnormal deteriorations in cognitive performance.
