DNA repair deficiency biomarkers and the 70-gene ultra-high risk signature as predictors of veliparib/carboplatin response in the I-SPY 2 breast cancer trial. [PDF]

open access: yes, 2017
Veliparib combined with carboplatin (VC) was an experimental regimen evaluated in the biomarker-rich neoadjuvant I-SPY 2 trial for breast cancer. VC showed improved efficacy in the triple negative signature.

The Role of Nicotinamide in Cancer Chemoprevention and Therapy

open access: yesBiomolecules, 2020
Nicotinamide (NAM) is a water-soluble form of Vitamin B3 (niacin) and a precursor of nicotinamide-adenine dinucleotide (NAD+) which regulates cellular energy metabolism.

PAR recognition by PARP1 regulates DNA‐dependent activities and independently stimulates catalytic activity of PARP1 [PDF]

open access: yesThe FEBS Journal, 2021
Poly(ADP‐ribosyl)ation is predominantly catalyzed by Poly(ADP‐ribose) polymerase 1 (PARP1) in response to DNA damage, mediating the DNA repair process to maintain genomic integrity. Single‐strand (SSB) and double‐strand (DSB) DNA breaks are bona fide stimulators of PARP1 activity. However, PAR‐mediated PARP1 regulation remains unexplored.

UBQLN4 Represses Homologous Recombination and Is Overexpressed in Aggressive Tumors [PDF]

open access: yes, 2020
Genomic instability can be a hallmark of both human genetic disease and cancer. We identify a deleterious UBQLN4 mutation in families with an autosomal recessive syndrome reminiscent of genome instability disorders.

PARP1-Erk synergism in proliferating cells [PDF]

open access: yesOncotarget, 2018
A synergism between PARP1 and phosphorylated Erk mediating IEG (immediate early gene) expression has been recently reported in cerebral neurons and cardiomyocytes. Stimulation induced PARP-Erk synergism was required for IEG expression underlying synaptic plasticity and long-term memory acquisition during learning.

Abstract 3709: PARP1 expression (PARP1expr) drives synergy between PARP1 inhibitors (PARP1-Is) and trabectedin (TR)

open access: yesCancer Research, 2016
Abstract Purpose of study. An attractive strategy to improve antitumor treatments is to inflict cytotoxic DNA damage with chemotherapy, and then impede DNA repair by molecular targeting. TR is a new drug characterized by a peculiar mechanism of action: TR traps DNA repair machinery leading to DNA damage, particularly in BRCA1/2-deficient

Role of poly(ADP-ribose) polymerase-1 in regulating human islet cell differentiation

open access: yesScientific Reports, 2022
Poly(ADP-ribose) polymerase-1 (PARP1), a fundamental DNA repair enzyme, is known to regulate β cell death, replication, and insulin secretion. PARP1 knockout (KO) mice are resistant to diabetes, while PARP1 overactivation contributes to β cell death ...

Dynamic Interplay between Cockayne Syndrome Protein B and Poly(ADP-Ribose) Polymerase 1 during Oxidative DNA Damage Repair

open access: yesBiomedicines, 2022
Oxidative stress contributes to numerous diseases, including cancer. CSB is an ATP-dependent chromatin remodeler critical for oxidative stress relief. PARP1 is the major sensor for DNA breaks and fundamental for efficient single-strand break repair.

PARP inhibitors affect growth, survival and radiation susceptibility of human alveolar and embryonal rhabdomyosarcoma cell lines [PDF]

open access: yes, 2018
PARP inhibitors (PARPi) are used in a wide range of human solid tumours but a limited evidence is reported in rhabdomyosarcoma (RMS), the most frequent childhood soft-tissue sarcoma.
