Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

Hydrolysis of 6-alkyl penicillins catalysed by ?-lactamase I from Bacillus cereus and by hydroxide ion [PDF]

open access: yes, 1988
Second-order rate constants for the hydroxide ion-catalysed hydrolysis of 6-alkyl penicillins are independent of the length of the alkyl side-chain and replacement of the amido by an amino group decreases the susceptibility to nuclephilic attack on 6 ...

Identifying transcription factors controlling the basal expression of human MRP4 highlights a substantial role for Sp1

open access: yesFEBS Open Bio, EarlyView.
The MRP4 transporter exports several drugs and signaling molecules. Here, we identified key promoter elements regulating basal MRP4 expression. Using reporter assays, we defined a conserved region with essential Sp1 and contributory Ets sites, which controlled basal MRP4 expression.

Derivation and characterization of retinal pigment epithelium from urine‐derived iPSCs

open access: yesFEBS Open Bio, EarlyView.
Age‐related macular degeneration causes vision loss via RPE dysfunction and loss. Traditional iPSC therapies rely on invasive biopsies, limiting scalability. Here, we utilize urine‐derived stem cells as an accessible source to generate u‐iPSCs, successfully differentiated into pigmented RPE. This “Urine‐to‐Retina” platform provides a promising path for

Group A Streptococcal Pharyngeal Infections: Diagnosis and Rational Antibacterial Therapy

open access: yesАнтибиотики и Химиотерапия, 2020
The problem of acute tonsillitis caused by Group A Beta-Hemolytic Streptococcus (GABHS) still remains relevant. This article provides data showing the revival of highly virulent GABHS infection and an increase in the incidence of complications (acute ...

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

Antagonism of the Antibacterial Action of Some Penicillins by Other Penicillins and Cephalosporins [PDF]

open access: yes, 1974
A B STRA CT There are many examples of two peni-cillins acting synergistically, usually by one competitively inhibiting 8-lactamase, thus protecting the other from inactivation. There are few reports on penicillins an-tagonizing each other. Eight strains

Pharmacological inhibition of the PERK pathway modulates hepatocellular carcinoma growth and immune signaling

open access: yesFEBS Open Bio, EarlyView.
Pharmacological inhibition of PERK in a DEN‐induced mouse model of liver cancer does not reduce tumor burden but alters cellular stress signaling. Despite blocking PERK activity, downstream stress responses, including CHOP expression, remain active, suggesting compensatory mechanisms within the unfolded protein response that may influence tumor ...

Tolerability of imipenem in patients with immediate hypersensitivity to penicillins [PDF]

open access: yes, 2006
Background Administration of imipenem/cilastatin to patients with IgE-mediated hypersensitivity to penicillins who might benefit from this treatment is usually avoided because of a 47.4% rate of cross-reactivity demonstrated in a single study on the ...
