On the Persistence of Particles [PDF]

open access: yesFoundations of Physics, 2005
This paper is about the metaphysical debate whether objects persist over time by the selfsame object existing at different times (nowadays called `endurance' by metaphysicians), or by different temporal parts, or stages, existing at different times (called ` perdurance').

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Metronidazole-tolerant Porphyromonas gingivalis persisters dampen cytokine expression in HGECs

open access: yes, 2019
Objectives: Our group has recently demonstrated the existence of metronidazole-tolerant Porphyromonas gingivalis persisters. This study further investigated the effects of P.

Single-cell Raman spectroscopy identifies E. coli persisters and their functionalities

open access: yes, 2021
Objectives: Microbial persisters are the featured tiny sub-population of microorganisms highly tolerant to multiple antimicrobials. Currently, studies on persisters remain challenging owing to technical limitations.

Cell‐cycle‐specific lesion evolution rather than inhibition of double‐strand‐break repair underpins cisplatin radiosensitization

open access: yesMolecular Oncology, EarlyView.
We analyze cisplatin–DNA adducts (CDAs) and double‐strand breaks (DSBs) in a cell‐cycle‐dependent manner. We find that CDAs form similarly across all cell cycle phases. DSBs arise only in S‐phase. CDAs might not directly impair DSB repair, but S‐phase DSB lesions evolve in the presence of CDAs and disrupt repair in G2, also causing radiosensitization ...

Specific Enrichment and Proteomics Analysis of Escherichia coli Persisters from Rifampin Pretreatment

open access: yes, 2018
Bacterial persisters, a dormant and multidrug tolerant subpopulation that are able to resuscitate after antibiotic treatment, have recently received considerable attention as a major cause of relapse of various infectious diseases in the clinic. However,

Hijacking emergency granulopoiesis: Neutrophil ontogeny and reprogramming in cancer

open access: yesMolecular Oncology, EarlyView.
Neutrophils are highly plastic innate immune cells; their functions in cancer extend beyond the tumour microenvironment. This Review summarises current understanding of neutrophil maturation and heterogeneity and highlights tumour‐induced granulopoiesis as a systemic programme that expands immature, immunosuppressive neutrophils via tumour‐derived ...

Persistent Mayer homology and persistent Mayer Laplacian

open access: yesFoundations of Data Science
In algebraic topology, the differential (i.e., boundary operator) typically satisfies $d^{2}=0$. However, the generalized differential $d^{N}=0$ for an integer $N\geq 2$ has been studied in terms of Mayer homology on $N$-chain complexes for more than eighty years. We introduce Mayer Laplacians on $N$-chain complexes.

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...
