Transfer of persulfide sulfur from thiocystine to rhodanese
Biochimica et Biophysica Acta (BBA) - Enzymology, 1979THiocystine (bis-[2-amino-2-carboxyethyl]trisulfide) is a natural substrate for rhodanese (thiosulfate:cyanide sulfurtransferase, EC 2.8.1.1). Analogs of thiocystine were prepared by eliminating the carboxyl or amino group or by lengthening the carbon chain.

Persulfide Signaling in Stress-Initiated Calmodulin Kinase Response
Antioxidants & Redox Signaling, 2020Significance: Calcium ion (Ca2+)/calmodulin (CaM)-dependent protein kinases (CaMKs) are activated by phosphorylation of a crucial threonine residue either by itself (CaMKII) or by upstream kinases, CaMK kinases (CaMKKs) (CaMKI and CaMKIV). CaMKs, present in most mammalian tissues, can phosphorylate many downstream targets, thereby regulating numerous ...

Chem. Commun., 2002
In support of the key features of sulfur transfer in the proposed mechanisms of 4-thiouridine generation, the enzyme ThiI can turn over only once in the absence of reductants of disulfide bonds, and Cys-456 of ThiI receives the sulfur transferred from the persulfide group of the sulfurtransferase IscS.

Measurement of Protein Persulfidation: Improved Tag-Switch Method
2019Hydrogen sulfide (H2S) is an endogenously produced signaling gasotransmitter, generated by the enzymes cystathionine γ-lyase, cystathionine β-synthase, and 3-mercaptopyruvate sulfurtransferase. The involvement of H2S in numerous physiological, as well as pathophysiological conditions, was established over the past decade. However, the exact mechanism(s)

Trafficking in persulfides: delivering sulfur in biosynthetic pathways
Nature Chemical Biology, 2006The presence of sulfur in cofactors has been appreciated for over a century, but the trafficking and delivery of sulfur to cofactors and nucleosides is still not fully understood. In the last decade, great strides have been made toward understanding those processes and the enzymes that conduct them, including cysteine desulfurases and rhodanese ...

Protein Persulfidation Protects Against Aging
2019For couple of decades aging is believed to be a consequence of damages accumulated by increased production of reactive oxygen species (ROS). Opposite to that, dietary restriction attempts to extend life have been linked to increased H2S production. We hypothesized that reacting with sulfenic acids (formed by ROS) H2S would rescue cysteine residue by ...
