Ageing acts as a double‐edged sword in cancer. In the elderly, open chromatin, immunosenescence, and chronic inflammation drive SASP (IL‐6, MMPs), MDSC accumulation and T‐cell suppression, fostering tumor‐promoting microenvironments and limited therapeutic benefit.

Lrp4+ astrocytes, uniquely vessel‐associated and GFAP−, are enriched in the pia. They support laminin‐α5+ basement membrane assembly and regulate vessel diameter and blood flow. They are reduced in 5xFAD brains, and their ablation worsens both vascular and Aβ pathology.
