Mammalian phospholipase C

2003
Publisher Summary This chapter provides an overview of mammalian phospholipase C. The hydrolysis of cellular phospholipids leads to the formation of various bioactive lipid mediators, acting either as extracellular signaling molecules or as intracellular second messengers. A well-known second messenger-forming system involving phospholipid hydrolysis

Purification of Phospholipase C β and Phospholipase C ε from Sf9 Cells

2003
Phosphatidylinositol-specific phospholipase C (PLC) enzymes catalyze hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) generating diacylglycerol (DAG) and inositol 1,4,5-triphosphate (IP(3)). The PLC beta isoforms of PLCs are activated by G proteins after hormone or neurotransmitter stimulation of G protein-coupled receptors (GPCR).

[22] Bioluminescent assay for lipase, phospholipase A2, and phospholipase C

1981
Publisher Summary This chapter describes the bioluminescent assay for lipase, phospholipase A 2 , and phospholipase C. The dim mutant M17 of the marine luminous bacterium Beneckea harveyi MAV #392 emits light with either exogenous aldehydes or fatty acids.

Protein kinase C‐dependent stimulation of phospholipase D in phospholipase C‐treated fibroblasts

Lipids, 1993
AbstractTreatment of [14C]choline‐ or [14C]ethanolamine‐labeled NIH 3T3 fibroblasts withBacillus cereus phosphatidylcholine‐specific phospholipase C (PLC) enhanced phospholipase D (PLD)‐mediated hydrolysis of the respective14C‐labeled phospholipids. PLD activity was stimulated by 1.5 U/mL of POLC and by 100 nM of the protein kinase C (PKC) activator ...

[49] Properties of phospholipase C isozymes

1991
Publisher Summary Phospholipase C (PLC)-catalyzed hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP 2 ) has been proposed to be a crucial step in the cellular response to calcium-mobilizing hormones and neurotransmitters. Numerous attempts to isolate PLC from various tissues have been made and as a result, various kinds of PLC have been ...

Phospholipase C-γ1 in tumor progression

Clinical & Experimental Metastasis, 2003
The vast majority of cancer morbidity and mortality arises from tumor progression beyond the primary tumor site. Unfortunately, most therapies are not effective for advanced stage disease with regional extension or distant metastases. Thus, new treatments are needed to target rate limiting steps in tumor progression.

Regulation of phospholipase C isozymes: activation of phospholipase C-γ in the absence of tyrosine-phosphorylation

Chemistry and Physics of Lipids, 1999
Activation of PLC-gamma isozymes in response to various agonists involves tyrosine phosphorylation of the effector enzymes. Recent evidence indicates that PLC-gamma isozymes are additionally activated by phosphatidic acid, phosphatidylinositol 3,4,5-trisphosphate and arachidonic acid in the absence of PLC-gamma tyrosine phosphorylation.

Phospholipase C- 1: A Phospholipase and Guanine Nucleotide Exchange Factor

Molecular Interventions, 2002
Although phospholipase C-gamma (PLC-gamma) participates in cellular mitogenesis, evidence indicates that the catalytic activity of PLC-gamma (to hydrolyze certain phosphoinositides) is nonessential to the process. So how is it that PLC-gamma is necessary but its lipase activity is not?

REGULATION OF EUKARYOTIC PHOSPHATIDYLINOSITOL-SPECIFIC PHOSPHOLIPASE C AND PHOSPHOLIPASE D

Annual Review of Biochemistry, 1997
This review focuses on two phospholipase activities involved in eukaryotic signal transduction. The action of the phosphatidylinositol-specific phospholipase C enzymes produces two well-characterized second messengers, inositol 1,4,5-trisphosphate and diacylglycerol.
