Codon pair optimization (CPO): a software tool for synthetic gene design based on codon pair bias to improve the expression of recombinant proteins in Pichia pastoris

open access: yesMicrobial Cell Factories, 2021
Background Codon optimization is a common method to improve protein expression levels in Pichia pastoris and the current strategy is to replace rare codons with preferred codons to match the codon usage bias.

A multi-model predictive control method for the Pichia pastoris fermentation process based on relative error weighting algorithm

open access: yesAlexandria Engineering Journal, 2022
The Pichia pastoris fermentation process is with highly nonlinear and time-varying and strong coupling characteristics, traditional control methods such as classic PID cannot satisfy the actual needs of the fermentation process control.

Cloning and Expression of Yak Active Chymosin in [PDF]

open access: yesAsian-Australasian Journal of Animal Sciences, 2016
Rennet, a complex of enzymes found in the stomachs of ruminants, is an important component for cheese production. In our study, we described that yak chymosin gene recombinant Pichia pastoris strain could serve as a novel source for rennet production ...

An improved CRISPRi system in Pichia pastoris

open access: yesSynthetic and Systems Biotechnology, 2023
CRISPR interference (CRISPRi) has been developed and widely used for gene repression in various hosts. Here we report an improved CRISPRi system in Pichia pastoris by fusing dCas9 with endogenous transcriptional repressor domains. The CRISPRi system shows strong repression of eGFP, with the highest efficiency of 85%.

High-yield overproduction and purification of human aquaporins from Pichia pastoris

open access: yesSTAR Protocols, 2022
Summary: Aquaporins (AQPs) are membrane-bound water channels that play crucial roles in maintaining the water homeostasis of the human body. Here, we present a protocol for high-yield recombinant expression of human AQPs in the methylotropic yeast Pichia

Expression of an anti-CD33 single-chain antibody by Pichia pastoris [PDF]

open access: yes, 2005
CD33 is a cell surface glycoprotein expressed on cells of myelomonocytic lineage, leukaemic cells, but not haematopoietic stem cells. By virtue of its expression pattern, CD33 has become a popular target for new immunotherapeutic approaches to treat ...
