Dynamic Feeding for Pichia pastoris [PDF]

open access: yes, 2022
The knowledge of certain strain-specific parameters of recombinant Pichia pastoris strains is required to be able to set up a feeding regime for fed-batch cultivations. These parameters are commonly determined either by time-consuming and labor-intensive continuous cultivations or by several, consecutive fed-batch cultivations. Here, we describe a fast

Expression of lignocellulolytic enzymes in Pichia pastoris [PDF]

open access: yesMicrobial Cell Factories, 2012
Background Sustainable utilization of plant biomass as renewable source for fuels and chemical building blocks requires a complex mixture of diverse enzymes, including hydrolases which comprise the largest class of lignocellulolytic enzymes.

Transcriptomics and Genomics in Pichia pastoris [PDF]

open access: yes, 2010

CLONING A FOREIGN GENE IN THE YEAST PICHIA PASTORIS [PDF]

open access: yes, 2022
The S gene encoding the hepatitis B surface antigen was cloned into the yeast Pichia pastoris using the pPIC9 transfer vector. The recombinant plasmid pPIC9-S was treated with the restriction enzyme SacI and inserted into the yeast Pichia pastoris via ...

Cloning and Expression of Yak Active Chymosin in [PDF]

open access: yesAsian-Australasian Journal of Animal Sciences, 2016
Rennet, a complex of enzymes found in the stomachs of ruminants, is an important component for cheese production. In our study, we described that yak chymosin gene recombinant Pichia pastoris strain could serve as a novel source for rennet production ...

An improved CRISPRi system in Pichia pastoris

open access: yesSynthetic and Systems Biotechnology, 2023
CRISPR interference (CRISPRi) has been developed and widely used for gene repression in various hosts. Here we report an improved CRISPRi system in Pichia pastoris by fusing dCas9 with endogenous transcriptional repressor domains. The CRISPRi system shows strong repression of eGFP, with the highest efficiency of 85%.

High-yield overproduction and purification of human aquaporins from Pichia pastoris

open access: yesSTAR Protocols, 2022
Summary: Aquaporins (AQPs) are membrane-bound water channels that play crucial roles in maintaining the water homeostasis of the human body. Here, we present a protocol for high-yield recombinant expression of human AQPs in the methylotropic yeast Pichia

Expression of an anti-CD33 single-chain antibody by Pichia pastoris [PDF]

open access: yes, 2005
CD33 is a cell surface glycoprotein expressed on cells of myelomonocytic lineage, leukaemic cells, but not haematopoietic stem cells. By virtue of its expression pattern, CD33 has become a popular target for new immunotherapeutic approaches to treat ...

Autolysis of Pichia pastoris induced by cold [PDF]

open access: yesAMB Express, 2017
The production of recombinant biopharmaceutical proteins is a multi-billion dollar market. Protein recovery represents a major part of the production costs. Pichia pastoris is one of the microbial systems most used for the production of heterologous proteins.

Towards better understanding of an industrial cell factory : investigating the feasibility of real-time metabolic flux analysis in Pichia pastoris [PDF]

open access: yes, 2013
Background: Novel analytical tools, which shorten the long and costly development cycles of biopharmaceuticals are essential. Metabolic flux analysis (MFA) shows great promise in improving our understanding of the metabolism of cell factories in ...
