Frequency and spectrum of PIK3CA somatic mutations in breast cancer [PDF]

open access: yesBreast Cancer Research, 2020
Purpose The therascreen PIK3CA mutation assay and the alpha-specific PI3K inhibitor alpelisib are FDA-approved for identifying and treating patients with advanced PIK3CA-mutated (PIK3CAmut) breast cancer (BC).

PIK3CA in Colorectal Cancer [PDF]

open access: yesFrontiers in Oncology, 2014
PIK3CA, the catalytic subunit of PI3K, is mutated in many different tumors, including colorectal cancer (CRC). Mutations of PIK3CA have been reported in 10-20% of CRC, about 80% of mutations found in two hot spots in exon 9 and exon 20. In RAS wild-type CRC, PIK3CA mutations have been associated with a worse clinical outcome and with a negative ...

Correlation between circulating cell‐free PIK3CA tumor DNA levels and treatment response in patients with PIK3CA‐mutated metastatic breast cancer [PDF]

open access: yesMolecular Oncology, 2018
Liquid biopsies focusing on the analysis of cell‐free circulating tumor DNA (ctDNA) may have important clinical implications for personalized medicine, including early detection of cancer, therapeutic guidance, and monitoring of recurrence.

Cancer-associated mutations in endometriosis reframe a benign disease through molecular oncology. [PDF]

open access: yesMol Oncol
This review aims to comprehensively analyse cancer‐associated somatic mutations (CAMs) present in endometriotic lesions, emphasizing their biological roles, spatial distribution and implications for translational applications in medicine. By contextualizing a benign state within a genomic framework, this analysis seeks to establish its value as a ...

Meme kanserinde PIK3CA [PDF]

open access: yes, 2018
Aim: In breast cancer (BCa) the frequency of PI3K signaling pathway and TP53 gene mutations are high and have role cancer development. The aim of the present study was to assess PIK3CA and TP53 mutations and PIK3R1, PTEN and SALL4 gene expression analysis and correlation with clinical parameters. Methods: PIK3CA and TP53 gene mutations in BCa samples ...

Natural Product Toosendanin Suppresses the Malignant Development of Skin Melanoma by Targeting BNC2 for Degradation. [PDF]

open access: yesAdv Sci (Weinh)
BNC2 exhibits context‐dependent opposing functions across multiple cancer types. This study reveals BNC2 as an oncogenic driver of melanoma proliferation and metastasis through transcriptional activation of PIK3CA. The natural compound TSN simultaneously degrades BNC2 and its oncogenic partner SMAD3 via CRBN‐dependent ubiquitination.

Aspirin promotes RSL3-induced ferroptosis by suppressing mTOR/SREBP-1/SCD1-mediated lipogenesis in PIK3CA-mutatnt colorectal cancer

open access: yesRedox Biology, 2022
Ferroptosis, a new form of regulated cell death triggered by the iron-dependent peroxidation of phospholipids, is associated with cellular metabolism, redox homeostasis, and various signaling pathways related to cancer.

Allosteric PI3Kα Inhibition Overcomes On-target Resistance to Orthosteric Inhibitors Mediated by Secondary PIK3CA Mutations

open access: yesCancer Discovery, 2023
Analyses of patients with advanced breast cancer resistant to orthosteric PI3Kα inhibitors revealed that secondary resistance mutations in PIK3CA alter the inhibitor binding pocket, which can be overcome by use of the allosteric PI3Kα inhibitor RLY-2608.

Мутации в гене PIK3CA при раке молочной железы с низкой экспрессией белка HER2 / neu [PDF]

open access: yes, 2022
Introduction. Disturbances in the PI3K-dependent (PI3K – phosphoinositide 3‑kinase) cascade are characteristic of all types of breast cancer. In particular, 30–40 % of patients with advanced / metastatic hormone-positive HER2‑negative (HER2 – human ...

PIK3CA and CCM mutations fuel cavernomas through a cancer-like mechanism

open access: yesNature, 2021
Vascular malformations are thought to be monogenic disorders that result in dysregulated growth of blood vessels. In the brain, cerebral cavernous malformations (CCMs) arise owing to inactivation of the endothelial CCM protein complex, which is required ...
