Optimizing the ER Cutoff in HER2‐Positive Breast Cancer: ER ≥ 50% Predicts Low pCR Rates and Resistance to Antibody‐Drug Conjugates in the Neoadjuvant Setting

open access: yesCancer Medicine, Volume 15, Issue 10, October 2026.
ABSTRACT Background Estrogen receptor (ER)‐positive/HER2‐positive breast cancer demonstrates significantly lower rates of pathological complete response (pCR) to neoadjuvant HER2‐targeted therapies compared to ER‐negative/HER2‐positive disease. However, the optimal ER positivity cutoff for clinically meaningful patient stratification remains undefined.

Next‐Generation Sequencing‐Based Molecular Profiling and Management of Advanced Solid Tumors: A Real‐World Experience in a Community‐Based Academic Cancer Center

open access: yesCancer Medicine, Volume 15, Issue 10, October 2026.
ABSTRACT Background Advances in molecular and cell biology have led to a paradigm shift in cancer management. We implemented next‐generation sequencing (NGS) at our community‐based academic cancer center over a decade ago. We sought to optimize the use of genomics and understand its impact in identifying actionable genomic alterations to determine ...

Anti‐Inflammatory Effects of Vicatia thibetica De Boiss. in Rheumatoid Arthritis and Associated Multi‐Pathway Modulation In Vivo

open access: yesChemistry &Biodiversity, Volume 23, Issue 10, October 2026.
Vicatia thibetica de Boiss. (VTDB) extracts and fractions were screened for anti‐rheumatoid arthritis activity via UHPLC‐MS, network pharmacology, and CIA rat models. VTDB‐nBuOH stood out, regulating PI3K/Akt, JAK2/STAT3, macrophage polarization, and gut microbiota to relieve joint inflammation, with proteomics verifying its multi‐pathway regulatory ...

Integrated Phytochemical, Immunological, and Network Pharmacology Investigation of Anti‐Inflammatory Lignans From Zanthoxylum armatum Fruits

open access: yesChemistry &Biodiversity, Volume 23, Issue 10, October 2026.
Bioassay‐guided fractionation of Zanthoxylum armatum fruits identifies three bioactive lignans—sesamin, fargesin, and eudesmin. The identified lignans attenuate CpG‐induced dendritic‐cell activation by reducing IL‐12 expression and CD80. Network pharmacology and molecular docking suggest involvement of multiple inflammatory signalling pathways ...

Systems Pharmacology and Multitarget Modulation of Letrozole–HFD‐Induced Polycystic Ovarian Syndrome by Fruit Pulp of Prunus armeniaca

open access: yesChemistry &Biodiversity, Volume 23, Issue 10, October 2026.
Integrated systems pharmacology and in vivo evaluation reveal that hydroalcoholic extract of fruit pulp of Prunus armeniaca (HAE of FPPA) ameliorates letrozole–HFD‐induced PCOS through multitarget modulation. Network analysis identifies 105 shared targets enriched in PI3K/Akt, MAPK, HIF‐1, and inflammatory pathways, while treatment improves hormonal ...
