Stage‐Specific H3K14 and H3K23 Succinylation Orchestrates Insect Metamorphosis and Oogenesis

open access: yesAdvanced Science, EarlyView.
Stage‐specific succinylation of histone H3 at lysine 14 and 23 differentially controls insect metamorphosis and oogenesis through distinct GPCR‐PKC signaling cascades, revealing an evolutionarily conserved epigenetic mechanism that coordinates key life‐history traits.

Polyphenol compounds and PKC signaling [PDF]

open access: yesBiochimica et Biophysica Acta (BBA) - General Subjects, 2016
Naturally occurring polyphenols found in food sources provide huge health benefits. Several polyphenolic compounds are implicated in the prevention of disease states, such as cancer. One of the mechanisms by which polyphenols exert their biological actions is by interfering in the protein kinase C (PKC) signaling pathways.

mTORC2 Phosphorylation of GSDME‐N Drives Cullin4B‐Mediated Proteasomal Degradation to Suppress Pyroptosis and Confer Radioresistance in Small Cell Lung Cancer

open access: yesAdvanced Science, EarlyView.
Radioresistance severely limits the efficacy of therapies for small cell lung cancer (SCLC). This study reveals a novel mechanism of resistance driven by the active suppression of pyroptosis. Specifically, the mTORC2 complex directly phosphorylates GSDME‐N and promotes its CUL4B‐mediated ubiquitination and proteasomal degradation.

A Novel Neuroprotective Mechanism of Riluzole: Direct Inhibition of Protein Kinase C

open access: yesNeurobiology of Disease, 2000
In addition to its antiexcitotoxic action, the anti-amyotrophic lateral sclerosis (ALS) neuroprotectant riluzole protects against nonexcitotoxic oxidative neuronal injury.

Imaging‐Guided Live Single‐Cell Lipid Profiling of Leader and Follower Cells During Collective Migration of Triple‐Negative Breast Cancer Cells

open access: yesAdvanced Science, EarlyView.
Integrated single‐cell mass spectrometry reveals distinct lipidomic signatures that differentiate leader and follower phenotypes in migratory breast cancer cells. Phenotype‐specific alterations across various lipid classes, including fatty acids and phosphatidylcholines, underpin functional heterogeneity.

ANXA2+ Small Extracellular Vesicles Drive Chemoresistance in Anaplastic Thyroid Cancer by Promoting XRCC5 Lactylation and Enhancing Non‐Homologous End‐Joining Repair

open access: yesAdvanced Science, EarlyView.
ANXA2+ sEVs promote cisplatin resistance in ATC by stabilizing the SRC/LDHA interaction, increasing LDHA phosphorylation and activity, and lactate production. Elevated lactate levels promote KAT5‐mediated lactylation of XRCC5 at lysine 265, enhancing its binding to XRCC6.

Differential and conditional activation of PKC-isoforms dictates cardiac adaptation during physiological to pathological hypertrophy. [PDF]

open access: yesPLoS ONE, 2014
A cardiac hypertrophy is defined as an increase in heart mass which may either be beneficial (physiological hypertrophy) or detrimental (pathological hypertrophy).

PKC and PKN in heart disease [PDF]

open access: yesJournal of Molecular and Cellular Cardiology, 2019
The protein kinase C (PKC) and closely related protein kinase N (PKN) families of serine/threonine protein kinases play crucial cellular roles. Both kinases belong to the AGC subfamily of protein kinases that also include the cAMP dependent protein kinase (PKA), protein kinase B (PKB/AKT), protein kinase G (PKG) and the ribosomal protein S6 kinase (S6K)

Penfluridol Triggers GSDME‐Mediated Immunogenic Pyroptosis to Potentiate Antitumor Immunotherapy

open access: yesAdvanced Science, EarlyView.
A high‐throughput screen of FDA‐approved antipsychotics identifies penfluridol as a potent pyroptosis inducer acting via direct TTI1 inhibition. This triggers TNFA‐NFKB signaling and caspase‐8/‐3‐dependent GSDME cleavage. The compound stimulates antitumor immunity alone and synergizes with anti‐PD‐1 therapy, while low TTI1 expression emerges as a ...

Retrospective analysis of protein kinase C-beta (PKC-β) expression in lymphoid malignancies and its association with survival in diffuse large B-cell lymphomas

open access: yesBiology Direct, 2007
Background Both mechanistic features and recent correlative findings suggest a potential role for protein kinase C-beta (PKC-β) in tumor pathogenesis, particularly in B-cell malignancies.
